Objective:This experiment aims to detect levels of mi R-328 and C/EBP α in the patients of chronic myelogenous luekemia(CML) blast phase(CML-BP) and chronic phase(CML-CP), in order to demonstrate abnormal expression of mi R- 328, C/EBP α, which can help us further understand the mechanics of CML leaps. Methods:1.28 patients with chronic myelogenous leukemia bone marrow specimens were divided into experimental group. There were 17 cases with chronic myelogenous leukemia chronic phase respectively(CML- CP), and 11 cases of acute phase of chronic myelogenous leukemia(CML- BP) in experimental group. While 22 cases with hyperplastic disease bone marrow specimens( 13 cases of the iron deficiency anemia, 9 cases of Idiopathic thrombocytopenic purpura) were treated as control group. Patients with other malignant tumor diseases were not including in control group.2.Western Blot were used to detect the expression of C/EBP α in marrow mononuclear cells. Then the datas were taken to statistical analysis.3.m RNA levels of mi R-328, C/EBP α were detected by real-time fluorescent quantitative PCR between groups. Then the datas were taken to statistical analysis. Results:1. Mi R- 328 expression level by real time- PCR, mi R-328 was expressed in CML-CP and control group while the expression was very low or non in CML-BP. CML-CP and CML-BP patients was lower than control group(P < 0.05). and mi R- 328 expression in CML-BP was lower than CMP-CP(P < 0.05).2. m RNA expression of C/EBP α by real time PCR, C/EBP α m RNA was expressed in each group. CML-CP and CML-BP was lower than control group(P < 0.05). No difference in m RNA expression of C/EBP α was found between CML-CP and CML-BP(P > 0.05);3. Protein expression of C/EBP α by Western blot, C/EBP α protein was expressed in CML-CP and control group while the expression was very low or non in CML-BP. It was lower in CML-CP and CML-BP period than the control group(P < 0.05). And C/EBP α expression in CML-BP was lowest of all(P < 0.05). Conclusion:1. Mi R- 328 expression decreased along with Chronic myelogenous leukemia chronic phase progression to Chronic myeloid leukemia blastic phase, which indicated mi R- 328 was associated with the progress of CML.2. Protein expression of C/EBP α decreased along with Chronic myelogenous leukemia chronic phase progression to Chronic myeloid leukemia blastic phase, while the m RNA expression levels of C / EBP α gene no significant change, which indicated expression C/EBP α was associated with the progress of CML.
