Radix Polygala is commonly used in traditional Chinese medicine, first recorded in "Shen Nong’s herbal classic", was listed as top grade, and was seen as drug to raising life. Tt stem from the dried root of Polygala tenuifolia willd or Polygala sibirica L, mainly in the northeast, north, northwest and central China and Sichuan; born in grassland, grassland slopes, scrub and forest under. Polygala taste bitter, acrid, warm, heart, kidney, lung, with efficacy of tranquilizer puzzle, traffic heart kidney, eliminating phlegm etc. For the heart kidney disharmony caused by insomnia, palpitation, forgetfulness trance, expectoration uncomfortable, sores, breast pain and other symptoms.This article based on a large number of references, the plant characteristics of Polygala tenuifolia, character, pharmacological action of a more comprehensive review, and then the experimental research on two aspects of Polygala tenuifolia.One aspect is the extraction and separation of tenuifolin system in Polygala tenuifolia, Firstly, strictly accordance with the 2010 Edition processing methods in "China Pharmacopoeia" contained to processe Radix Polygala with licorice gained Glycyrrhiza Radix Polygalae. Use orthogonal experiment to Choice the best technology of Glycyrrhiza Radix Polygalae for water extraction, After a comprehensive evaluation and analysis, A3B3C3 is the best extraction process the extraction of 90min, adding 12 times amount of water, decocting 3 times. After the water extraction and alcohol precipitation, and then on the macroporous resin, elution solvent and elution solvent dosage were selected, to determine the best process of macroporous resin to purification of total saponins in Polygala system. The final optimum conditions of AB8 macroporous adsorption resin for purification of total saponins of polygala, Elution solvent and dosage:distilled water,30% ethanol,70% ethanol, 3BV and eluted with a rate of 1BV/h, Collected the 70% ethanol elution. Using macroporous resin separate saponins of Polygala, its further acid treatment, extraction, silica gel column chromatography isolated tenuifolin. And the 6 kinds of different processed products of Radix Polygalae in tenuifolin were determined, the results are as follows:cinnabar> ginger> wine > liquorice> Crude Radix Polygalae>honey.On the other hand is the study of depression in female mice model for the extracted tenuifolin. Firstly, according to Karber’s method clear the LD50 value of tenuifolin on female mice, determination of tenuifolin dose; Then by isolated raised with chronic unpredictable mild stress (CUMS) method to establish female mice depression animal model, to observe the change of the biological characterization of the situation, according to the group the mice were intragastrically administered; then the behavior evaluation index and biochemical evaluation index were tested for the LSD-t by SPSS17.0 statistical software; Finally by the behavior evaluation index (changes of body weight、sucrose preference、open field test、swimming time) and biochemical evaluation index (MAO、ACTH、CORT)data statistical analysis, Experimental results show that in behavioral science:tenuifolin can significantly improve the appetite of depression in female mice and increase the happiness index of female mice, can improve the level of interest in female mice on the outside, can significantly reduce the time of despair, and enhance female mice in a dangerous state of desire to survive. In biochemical indexes:tenuifolin high and middle dose group can significantly reduce ACTH and CORT concentrations, and tenuifolin high, medium and low dose group can significantly reduce the content of MAO, tenuifolin can effectively improve the depression in female mice. At the same time, its mechanism may be related to the dysfunction of hypothalamus pituitary adrenal axis (HPA axis).This paper has always been the spirit of seeking perfection, pioneering and innovative spirit, strict inspection on the details of each experiment, Analysis on the pharmacological activity of tenuifolin processed Radix Polygalae in female mice of depression. The processing technology of Radix Polygalae, extraction and separation of saponins in Polygala tenuifolia for the studied chemical composition deeply, thus laied a foundation for further study of Radix Polygalae; Simultaneous determination of 6 kinds of different processed products of tenuifolin content, for the study of quality control of Radix Polygalae provides reliable scientific basis. More important is the first exploration of the use of tenuifolin study on depression model mice, and provide important reference value for the development of new drugs and clinical use of Polygala tenuifolia.
