Objective:To explore the protective effects of 1,25-dihydroxyvitamin D3 on renal by modulating the expression of transforming growth factor-beta 1 (TGF-β1), fibronection (FN) and vascular endothelial growth factor (VEGF) in diabetic nephropathy rats.Methods:Male SD rats were injected streptozotoein(STZ) to induce 36 diabetic models. The diabetic nephropathy rats were randomly divided into:TGF-β1 over-express peanut oil group;TGF-β1 over-express vitamin D3 group;TGF-β1 low-express peanut oil group;TGF-β1 low-express vitamin D3 group;TGF-β1 normal-express peanut oil group;TGF-β1 normal-express vitamin D3 group,each group had 6 rats. Both 1,25-dihydroxyvitamin D3 and peanut oil were given by lavage, kidney function, blood biochemical parameters were reevaluated after 37 days of treatment.The morphology of kidney tissues was tested by pathology. The expression of TGF-β1, FN, VEGF in kidney cortex were measured by immunohistochemistry in these groups.Results:The leves of cholesterol, triglyceride, creatinine,plasma glucose and 24-hour urinary protein were lower in vitamin D3 group than those in peanut oil group (P<0.05). The degree of renal fibrosis was associated with the levels of TGF-β1.Vitamin D3 decrease the fibrosis in diabetic kidney.The levels of TGF-β1 had significant difference in three peanut oil groups (P< 0.05); with the increasing levels of TGF-β1, the expression of FN and VEGF were increased.The expression of TGF-β1, FN and VEGF were higher in peanut oil group than those in vitamin D3 group (P<0.05) Conclusion: 1,25-dihydroxyvitamin D3 can protect diabetic nephropathy in rats by decreasing the fibrosis in kidney via inhibition of TGF-β1, FN and VEGF.
