ObjectiveIn current days, the tumor have become the first cause to the human’s health, especially the hepatocellular carcinoma (HCC). The morbidity and mortality of HCC in China even worldwide is increasing year by year. Therefore, looking for some new diagnostic and treatment-related molecules will contribute to the quickly and effectively diagnose and treat of HCC, which can improve the patients’curativeeffect and survival rate. Cyclooxygenase (COX) is very important in vivo, the study in recent years showed that COX-2 expressed higher in esophagus cancer and HCC, and have the relation with tumor angiogenesis. complement component 5a receptor (C5aR, CD88) is always used as the marker of leukocyte, which can bonding with the C5a and active the PI3K/A KT/mTOR and IL-6/ STAT3 pathways, which can promote the proliferation and anti-apoptosis of hepatic cells. The CD88 can chemotactic leucocyte and participate the inflammatory reaction and have the important role on the tumor invasion. The Previous studies showed that CD88 may promote the tumor angiogenesis and accelerate the invasion process. Because of the expression of COX-2 and CD88 in hepatocellular carcinoma tissues and the relationship with tumor invasion, this paper will explore the expression of the two molecules in hepatoma tissue and the relationship of them.Methods1. Specimen collectionCollect hepatocellular carcinoma tissues from the Department of Pathology of The First People’s Hospital of Xinxiang from January 2013 to April 2014, total 57 cases, which have complete data. Classified the specimens based on age, gender, tumor diameter, degree of differentiation and the state of lymph node metastasis. Some of the tissues stored in the liquid nitrogen, the others were fixed by 10% formalin solution and then embedded with paraffin and sect tissues for 4μm thickness.2. The expression of COX-2 and CD88 mRNA in the normal hepatic tissues and hepatocellular carcinoma tissues by RT-PCRExtract the total mRNA of the normal hepatic tissues and hepatocellular carcinoma tissues, detect the expression of COX-2 and CD88 mRNA by RT-PCR.3. Detect expression of COX-2 and CD88 protein with immuno-histochemical stainingSABC immunohistochemical staining method was used on the prepared tissue sections, observe and determine the expression site of COX-2 and CD88, calculate the expression rate and find if they are co-expression.4. Results judgeThe results of the RT-PCR were detected by the Image J software (grayscale values), capare the value of the objective band with internalreference to judge the expression of COX-2 and CD88 mRNA. Scored acording to the staining intensity and percentage of positive cells, then just the results with the cross product of staining intensity and percentage of positive cells.5. statistical treatmentMetering data are expressed by the mean±tandard deviation (x ±s), the data was treatmented with SPSS 17.0 statistical software. The average number of diverse compared using analysis of variance; Rate comparison with the χ2; Pearson correlation analysis test was used test the dependability, standards are a=0.05, in other words, P<0.05 means have significant statistical difference, P<0.01 means have a very significant statistical difference and P>0.05 means no statistical difference.Results1. The expression of COX-2 and CD88COX-2 and CD88 mRNA can be detected in the normal hepatic tissues and hepatocellular carcinoma tissues, and expressed higher in hepatocellular carcinoma tissues than in the normal hepatic tissues. The expression of COX-2 and CD88 mRNA raised up gradualy, there are no difference between low differentiated carcinoma and mid differentiated carcinoma, which are lower than the higher differentiated carcinoma (P< 0.05).2. The expression of COX-2 in HCC and the relationship with clinical pathological featuresYellow and brownish yellow particulate matter diffused in the cytoplasm of COX-2 positive cells, COX-2 positive rate in the HCC tissue was 73.68%. The expression of COX-2 protein in the HCC have no relation with tumor differentiation, tumor size, clinical stage and lymph node-independent metastasis (P>0.05).3. The expression of CD88 in HCC and the relationship with clinical pathological featuresThe expression of CD88 mainly located on the cell membrane, and some in the cytoplasm. The expression rate of CD88 protein in HCC tissue is 82.45%. The expression of CD88 protein in the HCC have no relation with tumor differentiation, tumor size, clinical stage and lymph node-independent metastasis (P>0.05).4. The correlation of the expression of COX-2 and CD88 in HCCIn COX-2 positive group of the HCC tissue, CD88 positive rate is 90.48%, significantly higher than COX-2-negative group (60%). The expression of COX-2 and CD88 protein showed correlation (r = 0.353, P<0.05).Conclusions1. The expression of COX-2 and CD88 mRNA in hepatocellular carcinoma tissues is more than that in the normal hepatic tissues, but the expression of COX-2 and CD88 protein in the HCC have no relation with tumor differentiation, tumor size, clinical stage and lymph node -independent metastasis2. There was a correlation between COX-2 and CD88 protein expression in the HCC.
