Objective:Observe the nourished Yin chemical fiber capsule on the cecal ligation and puncture (CLP) model of sepsis-induced in rats. Antithrombin Ⅲ (AT-Ⅲ)、 Platelet activating factor (PAF) impact. Provide a theoretical basis and new treatment methods for the treatment of sepsis medicine.Method:72 healthy male SD rats were randomly divided into four groups: Preliminary experiments group (The all cecal ligation group 8 only. The half cecal ligation group 8 only); Sham operation group 8 only.Model group(8h group.12h group.24h group 8 rats in each group); Chinese medicine treatment group (TCM group) (8h group.12h group.24h group 8 rats in each group) (1) Model of the steps of:After 24h fasting application taken by 10% chloral hydrate (0.4 ml/100 g) were anesthetized by intraperitoneal injection of the preparation of the animal model used. The rats were tied to the limbs and teeth fixed, supine located in the Operation table,Routine disinfection.Along the ventral midline abdominal skin incision of about 2 cm,, ①The all cecal ligation operation:Ligation all of the cecum.the No.18 needle puncture piercing of cecum 1-2 time ②The half cecal ligation operation:Ligation of the half cecum. No.7 needle Well versed in The piercing.③Sham operation:Separation of distal cecum and mesentery. Don’t do Cecal ligation. This study Model group and TCM group using Half a cecum ligation Making rat model of sepsis.(2) The research processing factors:Sham operation group and Model group Is 3 ml lavage physiological saline(After the operation 0.5h 12.5h Each time); TCM group Is 3 ml lavage nourished Yin chemical fiber capsule(After the operation 0.5h 12.5h Each time).(3) Experimental specimen collection and detection:Preliminary experiments group and Sham operation group Postoperative extracting tail blood 1 ml To blood culture. Preliminary experiments group 24h Live alive to kill rats An autopsy. Model group and TCM group At each point in time (8h.12h.24h) Each group alive kill, detection Antithrombin III(AT-III) and platelet activating factor (PAF).Result:1. Preliminary experiments group 8h Blood culture positive bacteria.24 h abdominal dissection examination see exudate; The all cecal ligation group 8 only 2 only death within 24 h. The half cecal ligation group 8 only. There is no death within 24 h. Sham operation group.8 h blood bacteria culture negative.24 h no death.2. Antithrombin-Ⅲ (AT-Ⅲ):8 h and 12 h TCM group and Model group Difference is not statistically significant(P>0.05).24h TCM group and Model group The difference is statistically significant(P<0.05).3. platelet activating factor (PAF):8 h and 12 h TCM group and Model group Difference is not statistically significant(P>0.05).24h TCM group and Model group The difference is statistically significant(P<0.05).Conclusion:1. The all cecal ligation operation and The half cecal ligation operation Can create the ideal model of sepsis. The all cecal ligation operation 24 h mortality 25%. The half cecal ligation operation 24 h no death. Sham operation group prove Cecum ligation is the essential condition of sepsis model success. The distal caecum and mesenteric separation Does not lead to the occurrence of sepsis.2. TCM group Antithrombin-Ⅲ (AT-Ⅲ) postoperative After 8 h began to rise. In 12 h,24 h test presents the rising trend. A statistically significant increase trend in 24 h The difference is statistically significant. TCM group platelet activating factor (PAF) postoperative After 8 h began to falling In 12 h,24 h test presents the falling trend A statistically significant increase trend in 24 h The difference is statistically significant.
