[Object] To analyze Mipu1’s structure and functions and to study the effects of Mipu1 on formation of foam cell from macrophages. [Methods and Results] The results of bioinformatic analysis showed that Mipu1 may be associated with oxidative stress and lipid metabolism, and there were two potential binding element of Mipu1 in CD36 promoter region via bioinformatic analysis websites and softwares.The result of real-time PCR and Western Blot showed that ox LDL induced the expression of Mipu1 and C D36 in animal level and cell level,which was in concentration and time depend ent manner. High performance liquid chromatography, fluorescence labeling method(Dil-ox LDL) and Oil Red O staining were used to observe the effects of Mipu1 on cholesterol accumulation in macrophages, the results showed that Mipu1 overexpression inhibited the accumulation of cholesterol in macrophages. Real-time PCR and Western Blot were used to explore the effect of Mipu1 overexpression on ABCA1, ABCG1, SR-B1 and CD36 expression, the results showed that Mipu1 overexpression inhibited the expression of CD36, however, it had no significant effect on expression of ABCA1, ABCG1,SR-B1.[Conclusion]Mipu1 can inhibition of macrophage foam cells induced by ox LDL,and its mechanism may be related to the regulation of expression of CD36.
