Purpose :The vascularized Mice Orthotopic Lung transplantation model has enormous research value in lung transplantation field. The establishment of this model faces significant challenge in microsurgical skills. Mice anatomical structure requires to be skillful, which needs to be done under microscope. Mice airways are so sensitive to outside stimulation with little endurance to chest-opening surgery, which adds difficult to airway management in airway intubation during and after surgery. As for the experimental equipment, because mice lung transplantation technique is still new and without establishment,there still needs reformed equipment during the surgery. Up to now, only a few countries are able to establish mice orthotopic lung transplantation model around the world. We will need to carry on following up researches based on this model. Therefore, we need to do related researches to the method of mice orthotopic lung transplantation model establishment.Methods :After surgical learning curve, in order to search for the best experiment condition, mice are divided into group of significant difference in receptor weight and group of insignificant difference in receptor weight,LPD group and Euro-Collins group, lung artery group and lung veins group,large diameter casing anastomosis group and small diameter casing anastomosis group, neap gas high respiratory frequency ventilation group and neap gas low respiratory frequency ventilation group,CT evaluation will be utilized to value transplanted lung image expression and to observe pathology change in transplanted lung tissue by HE staining.Results: It further improves model establishment method, which is to infuse donor’s lung by use LPD through lung artery and lung veins. Bronchoalveolar,lung veins and lung artery were cased by 20 G,22G,24 G tubes.The receptors and donors were ventilated by high-frequency ventilation method during the surgery(with respiratory of 110 times a minute and ventilation amount 0.5 ml).Conclusions:The Vascularized Mice Orthotopic Lung transplantation model can be established after being experienced of learning curve and be standardized. The establishment of mice orthotopic lung transplantation model can be optimized through weight matching of receptor, lung refusing method, lung protection, casing size and respiratory machine paraemeters.
