Objective: To develop the method with good selectivity and high sensitivity for Ibuprofen determination by the high performance liquid chromatogram tandem mass spectrometry(LC-MS/MS), to provide the technical support and application reference for rational use and clinical pharmacokinetics research.Methods:Based on the constitutional formula and speculative knowledge of Ibuprofen, the LC conditions were optimized, including the components of mobile phase, elution mode, column type and column temperature and so on. The methods of determination for Ibuprofen in matrix were developed and validated following the national guidelines. The data was collected and analyzed by software, and the data were calculated by Excel.Results:I. An LC-MS/MS method was developed and validated for the quantitative determination of Ibuprofen in rat plasma. A sample clean-up pretreatment by protein precipitation was used. The limit of quantification of this method is 10.0 ng/m L.II. An LC-MS/MS method was developed and validated for the quantitative determination of Ibuprofen in rat plasma.
