Objective: Hemin intraperitoneal injection after building Allen’s rat spinal cord injury model. Analysis the changes and differences of the behavior scores between different groups rats after SCI.To explore the regulations of neuroglobin(Ngb) expression and Ngb location in the rat spinal cord after Hemin intraperitoneal injection. And The protective effect of high expression of spinal cord Ngb induced by Hemin in SCI about spinal cord structure, cell level and pathological process. Try to discover a new research direction and provide reliable laboratory supports in SCI treatment.Methods:90 healthy male SD rats,(weight between 180g to 200g)provided by the Military Academy of Medical Sciences, and were randomly divided into three groups: control group(sham operation group), surgical group, Hemin treatment group(by intraperitoneal injection with Hemin after building SCI model). Control group lamina resection only, not to injury the spinal cord, and intraperitoneal injection with saline solution. Surgical group was built SCI model according to Allen’s method. Both control group and surgical group are intaperitoneal injection with saline solution; Hemin treatment group was intraperitoneal injection with the same amount of Hemin. Through the Basso, Beattie, Bresnahan locomotor rating (BBB) scale, hematoxylin-eosin(HE) staining, Nissl’s staining and immunohistochemical to evaluate the changes and differences of the three groups of rats in 1d,3d,7d,14d,28d,56d about movement function, spinal structure, content and distribution of Ngb expression in the spinal. All the date were analysed results with statistics software SPSS.Results:The results of the three groups at particular points in time as follows:(1) The BBB score:control group> Hemin treatment group> surgical group. (2) HE staining results:control spinal structure is clearly without bleeding, cavity, inflammatory cell infiltrate, cell edema and cell degeneration. Surgical group spinal tissue contain obvious bleeding, cavity, nerve tissue inflammatory cell infiltration, cell edema, cell degeneration and neuronophagia. The damage of Hemin treatment group is lighter than the surgical group with less amount of phenomena as is mentioned above. (3) Nissl’s staining results:(1)Nissl bodies of control group are normomorph around the nucleus in granular, and deeply color and more extensive distribution. (2)Surgical group neuron degeneration is heaviest. There is austenite dissolvein phenomenon addition to the cell edema, vacuolization and more shallow color, fewer Nissl bodies. (3)Hemin treatment groups showed better than the surgical group with edema of nerve cells and the decrease in the number of color becomes shallow. (4) Immunohistochemical results:1)The number of positive cells in the spinal cord tissue within 7days: Hemin treatment group> surgical group> control group.2)The number of positive cells in the spinal cord tissue is no significant difference among the three groups after 7days.Conclusion:This experiment intervene that the Allen’s SCI model of rat with Hemin intervention. To verify the expression induced by SCI and the high expression induced by Hemin. To explore and study the effect of high expression induced by the Hemin injection in SCI. And the results of this experiment indicate that Hemin can improve the resistance of spinal cord to SCI, include oxidative stress injury, inflammatory reaction and cell death apoptosis. The protective effect of Ngb has multiple pathways, and elements. We can increase the level of Ngb expression of spinal cord to improve its neuroprotective effect, and may provide reliable laboratory theoretical basis and a new ideas to treat the SCI.
