| Objective :To evaluate the changes of neuroglobin expression after spinal cord ischemia/reperfusion injury (SCI) in the rats.Methods :Fifty six adult Wistar rats (250~300g) were randomly divided into 6 groups, including sham operation group (n=6) and five operation groups (n=10). The spinal cord SCI model was established with a self-made device in the operation groups.The expression of NGBmRNA was detected by using in situ hybridization histochemistry analysis, and the expression of NGB was detected by using Western blotting in spinal cord tissue.Results :The expression of NGBmRNA was increased at 2h after SCII, and peaked at 48h,but appeared to reduce significantly after reperfusion for 72h. The NGBmRNA optical density level at 2h, 12h,24h ,48h and 72h groups was(0.26±0.05),(0.28±0.04),(0.30±0.05),(0.41±0.08) and (0.32±0.06),respectively and significantly higher than that incontrol groups(P<0.05).Conclusions :NGB was induced by SCII in the rats,and changed in a time-dependent manner. It was one of the endogenous nerve protective mechanisms. |
