| Objective: To investigate the expression of MCP-1 and the infiltration of mononclear phagocytes after the spinal cord injury in rat and the influence after hyperbaric oxygen therapy.Methods: Seventy-two adult male Sprague Dawley rats were randomly assigned into spinal cord injury group(group A), HBO treatment group(group B), and sham operation group(group C). Each groupw were divided into three subgroups 1dã€3dã€7d group, 8 rats in each group. In groups A and B, the SCI model was established with modified Allen’s method and the rats of group C served as control, which canal was opened without damage to spinal cord. Then group B underwent hyperbaric oxygen therapy after 6h post-operation. The other two groups didn’t accept hyperbaric oxygen therapy. Using BBB score assessed rates hind limbs motor function recovery after after postoperative. At 1dã€3dã€7d after operation, spinal cord tissues were collected and sections were made to do following examinations: RT-PCR method tested the expression of MCP-1 m RNA, Western blot method detected the secretion of MCP-1 protein. The expression of MCP-1 and ED-1positive cells at different time points were detected by immunohistochemistry method, HE staining observed spinal cord injury tissue morphology changes.Results: At different time points, the BBB score of group C was significantly higher than those of groups A and B(P < 0.05), and the BBB score of group B was significantly better than that of group A at 3 and 7days after operation(P < 0.05). Molecular biology examination, groups were all in the expression of MCP-1, at each time point A groups of MCP-1 expression were significantly higher than that of group C(p<0.05). At each time point, the expression of MCP-1 of A group was obviously higher than that of group B(p<0.05). Immunohistochemical staining, At each time points Aã€B groups of MCP-1 integral optical density(IOD) values are higher than C group(p<0.05). At 3dã€7d time point, the IOD values of MCP-1 in A group was significantly higher than that of group B(p<0.05). Each time point Aã€B group of ED-1 number of positive cells were higher than that of group C(P < 0.05). At 3dã€7d time point, the ED-1 positive cells were higher than group B(P < 0.05).HE staining observated SCI group of spinal cord tissue structure of disorder, inflammatory reaction on HBO treatment group obviously.Conclusions: Hyperbaric oxygen treatment can deduce the second inflammation action by inhibiting the level of MCP-1 expression, meliorating the degree of secondary injury after SCI and improving the neural function recovery. |
