Objective To investigate the puerarin of the sacral spinal cord injury(SSCI) lead to bladder detrusor dysfunction abnormalities therapeutic effect and its mechanism.Methods The 30 healthy adult female SD rats were randomly divided into three groups: control group, model group and treatment group,10 per group. The control group received no surgical treatment, the model group and treatment group were neurogenic bladder spinal cord transection model was prepared due to spinal cord injury. Control group and model group were injected with saline solution treatment group were injected rhynchophylline 5mg / kg. After four weeks, using urodynamic bladder dysfunction detected cases of isolated bladder detrusor strips electrophysiological indicators detect detrusor hyperreflexia case, sliced HE staining of pathological changes in the detrusor, immunoblot assay force urine muscle ICC C-kit protein expression cells, confocal microscopy ICC cell number changes, real-time PCR detection of ICC cells T-type calcium channel protein isoforms α1G gene expression levels.Results Compared with the control, the other two groups leak point pressure, bladder filling pressures were increased(P <0.01), the maximum bladder capacity is reduced(P <0.01), increased contraction frequency(P <0.01), the smallest decrease tension(P <0.01), C-kit protein expression increased(P<0.01), an increase in the number of ICC cells(P <0.01), α1G gene expression increased(P <0.01). Compared with the model group, the treatment group bladder leak point pressure, bladder filling pressures are decreasing(P <0.01), the maximum bladder capacity increased(P <0.01), lower contraction frequency(P <0.01), the smallest increase tension(P <0.01), C-kit protein expression were decreased(P <0.01), reducing the number of ICC cells(P <0.01) α1G gene expression was decreased(P <0.01). Detrusor tissue pathological changes: light microscope, the control group detrusor oval or fusiform cells, arranged in parallel between clear cell distribution. Detrusor fracture model group, cell disorder, uneven distribution, cell gap widened, elastic fibers decreased collagen fibers. Lightly stained cytoplasm, myocyte hypertrophy, there are a large number of neutrophil infiltration. Detrusor rare fracture treatment group, less collagen fibers and intercellular alleviate inflammatory cell infiltration.Conclusion Rhynchophylline by reducing the number of cells on the detrusor ICC, inhibition of T-type calcium channel subtypes α1G ICC cells to inhibit the expression of excited activity pacing, thereby reducing excessive detrusor hyperactivity that tends to normal detrusor excitability.
