Purpose:Explore the conditions and synthetic methods of N-acylsulfonamides and a-acyloxyenamides using ynamides in presence of metal-free. Analyze and confirm the structure of target product, preparate a series of N-acylsulfonamides and a-acyloxyenamides to test pharmacological activity. Method:1. Design and carry optimized reaction conditions which mainly base on the idea of organic chemistry methodology.2. Basic experimental operations follow the standard of pharmaceutical chemistry rule.3. Monitor the reaction by thin layer chromatography (TLC) and nuclear magnetic resonance (NMR) spectroscopy.4. Separate compound by column chromatography.5. Determine compound structure with nuclear magnetic resonance (NMR) spectroscopy, mass spectrometry and X-ray single crystal diffraction. Result:Get the best reaction conditions for synthesis of N-acylsulfonamides and a-acyloxyenamides by ynamides under the condition of metal-free. Obtain a series of N-acylsulfonamides and a-acyloxyenamides, and determine the exact structure of product with nuclear magnetic resonance (NMR) spectroscopy, mass spectrometry and X-ray single crystal diffraction. Conclusion:Synthesis a series of N-acylsulfonamides and a-acyloxyenamides by metal-free hydration and hydroacyloxylation reactions of ynamides. This is the best way to avoid the trace of metal in product, reduce the steps of purification. The novel reaction processes prepare a series of N-acylsulfonamides and a-acyloxyenamides with important drug structure, which is a simple, eco-friendly and high yield method.
