Objective: To observe the efficacy of Kiwi essence on theadenocarcinoma in mice and the effect of chemotherapy.Methods: Forty C57BL/6J mice bearing Lewis cells were randomized into five groups: Control group(normal saline), Kiwi essence group(Kiwi essence 180 mg/kg), Chemotherapy group(cisplatin and gemcitabine), Low-dose Kiwi essence combined chemotherapy group(Kiwi essence 60 mg/kg combined cisplatin and gemcitabine) and High-dose Kiwi essence combined chemotherapy group(180 mg/kg Kiwi essence combined cisplatin and gemcitabine). After inoculated with adenocarcinoma cells after 4 days, drug intervention were given in five groups of mice. Mice were sacrificed at day 20 after tumor inoculation.During the course of experiment, to observe the mice transplanted tumor growth. the metastasis nodules on lung surface were measured in every group. The lung tissue were observed by HE dyeing. The expressions of vascular endothelial growth factor(VEGF), Bcl-2 and Survivin were determined by immunohistochemistry method with image analyze system. The m RNA expressions of apoptosis-related genes(Survivin andBcl-2) and drug resistance genes(ERCC1 and RRM1) were detected by RT-PCR. The protein expression of ERCC1 and RRM1 were examined by western blot.Results:1. Mice transplanted tumor growth and metastasis situationThe weight and volume of transplantation tumor in the drug intervention groups were lower than the control group, the High-dose Kiwi essence combined chemotherapy group was lower than other drug group(P<0.01), The weight of subcutaneous tumor was(5.95±0.20)g,(5.06±0.45)g,(3.53±0.52)g,(3.18±0.93)g and(2.29±0.31)g in control group, Kiwi essence group, chemotherapy group, low-dose Kiwi essence combined chemotherapy group and high-dose Kiwi essence combined chemotherapy group respectively; The volume of subcutaneous tumor was(6.66±0.49) cm3,(5.55±0.56) cm3,(3.72±0.39) cm3,(3.41±0.48) cm3and(2.41±0.58) cm3 in every group respectively; The amount of metastasis nodules on lung surface was(14.6±2.6),(9.9±1.7),(9.3±1.8),(5.7±1.6) and(2.0±1.1) in every group respectively. The tumor inhibitory rate was 14.95%, 41.77%, 44.21% and 59.68% in Kiwi essence group,Chemotherapy group, Low-dose Kiwi essence combined chemotherapy group and High-dose Kiwi essence combined chemotherapy group respectively; The metastasis rate on lung surface was 32.19%, 36.3%,60.7% and 86.4% in Kiwi essence group, Chemotherapy group,Low-dose Kiwi essence combined chemotherapy group and High-dose Kiwi essence combined chemotherapy group respectively.2. The expression of VEGF in the transplanted tumor of miceThe expression of VEGF in the control group were higher obviously than the treatment groups, and the High-dose Kiwi essence combined chemotherapy group was lower than other drug group(P<0.01).The expression of VEGF were(150.97±7.06),(129.75±6.41),(111.29±9.89),(98.84±8.46) and(80.56±6.66) in control group, Kiwi essence group,chemotherapy group, low-dose Kiwi essence combined chemotherapy group and high-dose Kiwi essence combined chemotherapy group respectively.3. The expression of apoptosis-related genes(Survivin and Bcl-2) in the transplanted tumor of miceThe expression of Survivin and Bcl-2 in the control group were higher obviously than the treatment groups, and the High-dose Kiwi essence combined chemotherapy group was lower than other drug group(P<0.01), The m RNA of Bcl-2 were(101.59±8.15),(83.42±5.42),(68.65±2.47),(52.93±2.90) and(35.94±5.46) in control group, Kiwi essence group, chemotherapy group, low-dose Kiwi essence combined chemotherapy group and high-dose Kiwi essence combined chemotherapy group respectively; The m RNA of Survivin were(111.43±9.39),(92.41±6.98),(82.02±1.59),(62.02±3.12) and(53.07±2.67) in every group respectively, The expression of Bcl-2 were(148.98±9.61),(128.92±4.89),(106.45±7.66),(97.54±3.55) and(77.89±7.31) in every group respectively; The expression of Survivin were(158.88±8.11),(138.48±9.28),(125.85±8.92),(110.44±7.80) and(84.14±11.42) in every group respectively.4. The expression of drug resistance genes(ERCC1 and RRM1) in the transplanted tumor of miceThe expression of ERCC1 and RRM1 in the control group were higher obviously than the treatment groups, and the High-dose Kiwi essence combined chemotherapy group was lower than other drug group(P<0.01),The expression of ERCC1 m RNA were(104.62±4.50),(58.59±2.28),(99.45±4.45),(82.46±2.57) and(57.68±3.76)in control group, Kiwi essence group, chemotherapy group, low-dose Kiwi essence combined chemotherapy group and high-dose Kiwi essence combined chemotherapy group respectively; The expression of RRM1 m RNA were(102.30±5.50),(60.55±3.51),(98.84±4.44),(80.66±6.35) and(55.60±3.62) in every group respectively, The expression of ERCC1 protein were(86.29±2.84),(40.04±1.50),(82.66±2.68),(43.47±3.42) and(37.03±3.17) in every group respectively; The expression of RRM1 protein were(97.58±2.46),(38.36±2.15),(94.77±3.61),(57.09±1.68) and(33.29±4.20) in every group respectively.Conclusion:1. These indicated that Kiwi essence likely inhibits the growth and metastasis effectively of Lewis lung cancer in mice.2. The Kiwi essence likely increase sensitivity to chemotherapy effectively of Lewis lung cancer in mice, which is involved in the suppression of RRM1,ERCC1 expression.
