Objective:In this study,we established the model of vascular othel ial injury in rats.2 weeks after giving sappanwood ethyl acetate extract suspension by gavage,we detect the effect of the contents of soluble int ercellular adhesion molecule-1 (sICAM-1) in the serum of the rats, and observe pathological changes of rat vascular endothelial cells, so as to investigate the effect of sappanwood ethyl acetate extract on vascular e ndothelial protection.Methods:Wistar rats were randomly divided into:blank control gr oup, model control group,sappan group, simvastatin group,except for the blank control group,other groups were treated with 3% methionine diet for 6 weeks,the establishment of model of vascular endothelial damage. After the success of model building given sppan acetic ether group gav age concentration 73.6mg/ml sappan acetic acid ethyl ester extract suspe nsion 10ml/kg·D (crude drug 1.15g/kg·D) for 2 weeks, simvastatin grou p gavage simvastatin suspension 10ml/kg·D(equivalent to simvastatin 5.0 mg/kg·D) for 2 weeks,the blank control group and model control group were administrated by volume of sodium cellulose solution 2 weeks obs erved the general condition of the rats in each group were changed in the After 2 weeks,the pathological changes of blood vessel endothelium were observed, and the content of sICAM-1 in serum was detected by ELISA,and the data were analyzed by SPSS18.0 software.Results:1.Blank control group in thoracic aorta,vascular endothelia 1 cell surface integrity closely spaced; the model control group,simvasta tin group and sppan acetic ethyl group of vascular endometrial cell inju ry significantly. The damage degree Sppan acetic ethyl group and simva statin group compared with model control group is light.2.Compared wit h the blank control group, the serum levels of sICAM-lof model contro 1 group, sappan group and simvastatin group were significantly increase d,the difference is statistically significant (P<0.05);Compared with the model control group, the serum levels of sICAM-lof sappanwood ethyl acetate group and simvastatin group were significantly decreased.the diff erence is statistically significant (P>0.05). Compared with sappanwood e thyl acetate group, the serum sICAM-1 of simvastatin is decreased, but the difference was not statistically significant (p>0.05).Conclusion:1.Sappanwood ethyl acetate extract could reduce the d egree of endothelial damage of vascular endothelial injury model in rats. 2.The extract can reduce the serum sICAM-1 level of rats with vascul ar endothelial injury.
