| Objective:To eatablilsh the quality evaluation of roots of Foeniculum vulgare Mill, based on the Chinese Pharmacopeia (2010). Methods:Ethyl acetate extract part were isolated and pured by silica gel column chromatography, Sephadex LH-20 and recrystallization. The experiment was took the roots of Foeniculum vulgare Mill. form 10 areas in Xinjiang as the object of study, analyzed the volatile oil of roots of Foeniculum vulgare Mill, and carried on the quality evaluation though three parts to the research.The content of quality evaluation including:identification (character, microscopic identification, TLC identification), inspection (moisture, total ash and acid-insoluble ash, soluble extracts, heavy metals,arsenic salt), content determination and so on. Results:1.Five compounds were isolated from the ethyl acetate extraet part,they were identified as dillapiol and butyl isobutyl phthalate and linoleic acid and β-sitosterol and 3R,8S,9Z-falcarindiol. 2.Twenty-six components in the volatile oil of roots of Foeniculum vulgare Mill, from ten different habitats were identified. dillapiol and apiole and myristicin are its common, which account for 90.81%-96.18% and 0.54%-2.76% and 0.10%-3.33% of the total oils. 3. Properties and microscopic characteristics of roots of Foeniculum vulgare Mill.were studied and obtained six microscopic characteristics:catheters,cork cells,oil chamber, wood fiber,starch grains and parenchyma cells.It is the first time to establish a method by using thin layer chromatography(TLC) for simultaneous identification of dillapiole and β-sitosterol of roots of Foeniculum vulgare Mill..4. Result of routine examination: the limit of the contents of moisture,total ash,total insoluble ash,cold or hot water-soluble extracts.cold or hot alcohol-soluble extractive were 10.00%,10.00%,2.50%,30.00%, 45.00%,13.00% and 20.00%.5.The Gas Chromatography was established to determine the content of dillapiol of roots of Foeniculum vulgare Mill.. The relationship between the concentration and the peak area of dillapiol is good linear relation in 5.08μg~50.80μg, r=0.9999,an average recovery rate is 99.86%, RSD=1.20%. The results of content determination of dillapiol in roots of Foeniculum vulgare Mill. from different habitats is 106.1mg/100g~159.7mg/100g.6.The high performance liquid chromatography was established to determine the content of (3-sitosterol of roots of Foeniculum vulgare Mill.. The relationship between the concentration and the peak area of β-sitosterol is good linear relation in 0.52μg-12.61ug, r=0.9998,an average recovery rate is 99.97%,RSD=1.58%. The results of content determination of β-sitosterol in roots of Foeniculum vulgare Mill. from different habitats is 21.38mg/100g~29.93mg/100g. Conclusion:Established the quality evaluation of Foeniculum vulgare Mill. though identification,Inspection and determination,which supplies the modern method of quality evaluation for traditional Chinese drug with qualitation and quantitation analysis. |
