Objective:To observed the expression of A2B Adenosine Receptor in spleen and IL-6 in serum in EAE mice which is an animal model of multiple sclerosis,we discussed the therapeutic effect of aerobic exercise(treadmill) and explored its mechanisms in MS or EAE.Methods:60 C57BL/6 mice were randomly assigned to 3 groups:adjuvant control group(CG), EAE model group(MG) and aerobic exercise group(AG).The MG and AG consisted of mice immunized with MOG35-55 emulsified in CFA.The CG consisted of mice injected with PBS emulsified in CFA.Both of the CG and MG,the mice were conducted without any intervention.The mice in AG trained on day 3 post-immunization on the treadmill apparatus,6 days/week during 4 weeks at moderate intensity.Weight changes and clinical scores of each group were recorded and graded.All mice were sacrificed on day 30 post-immunization,the expression of A2B Adenosine Receptor in spleen were measured by immunohistochemistry, Interleukin-6 in serum were measured by ELISA.Results:1. The weight changes of the mice in each group:to compare mice in CG,mice in MG losted significant weight on day 15,20,25,30 post-immunization(P<0.05),to compare mice in MG,mice in AG gained significant weight on days 15 post-immunization(P<0.05).2. The clinical scores of the mice in each group:to compare mice in CG,the clinical scores of mice in MG were significant high(P<0.05),but mice trained with treadmill reduced clinical scores compared to model mice on day 20,25,30 post-immunization (P<0.05).3. EAE onsets of the mice in each group:there was no EAE in CG,to compare mice in MG,aerobic exercise showed obvious to delay EAE onset(P<0.05).4. The expression of A2B Adenosine Receptor in mice spleen:A2B Adenosine Receptor of mice in MG was significantly upregulated,compared with CG(P<0.05),the statistical lower level of A2B Adenosine Receptor of mice in AG than that in MG(P<0.05).5. The change of Interleukin-6 in mice serum:Interleukin-6 was upregulated in MG,to compare with CG(P<0.05),aerobic exercise caused a statistical lower level than that in MG(P<0.05).Conclusion:1. Aerobic exercise could delay the EAE onset time,ameliorate EAE score.2. The downregulation of A2B Adenosine Receptor and IL-6 after aerobic exercise could play a role of regulating inflammatory reaction in EAE.
