In order to obtain an ideal artificial cornea, the optical center of artificial cornea was modified through surface modification and doping modification.1、Highly hydrophilic and highly hydrophobic polymer were synthesized and characterized by IR, NMR, etc.surface modification of artificial cornea optical center which composed of polyvinyl alcohol(PVA) hydrogel,was carried out by those polymers.In order to confirm the influence of surface modification on the material properties, the article tests the property of modification material and raw material by the contact angle, moisture content, mechanical properties, visible light transmittance, swelling balance, etc.were measured.2、After synthesis, the optical center material was doped by the highly hydrophilic or highly hydrophobic polymer,In order to confirm the feasibility and practicality of the doping modification material,the properties between the doped polymer and undoped polymer were compared by the contact angle, moisture content, mechanical properties, the optical transmittance, swelling balance, etc.3、The biocompatibility of the modified material was evaluatedby in vitro toxicity testing, cell adhesion assay, intradermal reaction andhemolysis experiments. The results showed that:(1) The toxicity grade were 0 or 1 which meaned the material was nontoxic.(2) The surface of the material showed no cell binding, suggesting near-complete blockade of cell adhesion.(3) The intradermal reaction was minimal.(4) No obvious hemolysis phenomenon was observed. The experiments proved that the modified material has achieved national standard for medical material.4、The modified material and the scaffolds material were used to manufacture an artificial cornea. The animal experiment was carried out. After keratoprosthesis, ocular photoreception was detected in the experimental rabbits. Further exploring of the implantation was done by optical coherence tomography(OCT) and histopathology examination.
