Objective:At present, lung cancer is one of malignant tumors with the highest incidence and mortality in the world, and the morbidity and mortality increased year by year, and this trend is particularly obvious in China. Although the treatment continuous improvement and progress in the last 10 years, and the survival rate has increased, but the mortality remains high, five years total survival rate is still low, the fundamental reason is the low rate of early diagnosis and the cure rate is low. The study of single nucleotide polymorphism(SNP) and lung cancer susceptibility can provide a lot of helps on the screening of lung cancer susceptible population and early diagnosis and early treatment and prognosis. In this study, we selected two functional sites of rs4648127(C/T) and rs230496(A/G) of NF-κB1 intron and rs1948 of CHRNB4 gene exon,and study the correlation between SNP and lung cancer susceptibility.Methods:There are 353 patient with lung cancer and 379 able-bodieds who from First Affiliated Hospital,Soochow University include in this study. Extract their genomic DNA of peripheral blood samples, and using high resolution melting curve(HRM) technology for genotyping. The data were statistically analyzed with SPSS19.0 statistical analysis software. Cases and the control group of the genotype and allele frequency distribution differences with chi square test analysis of, and the association of these polymorphisms and lung cancer susceptibility with Logistic statistics, OR value and 95%Cl of lung cancer risk, and analyses were stratified according to age, sex, smoking and drinking.Results:The rs4648127 of NF-κB1 intron mutation gene CT and the susceptibility to lung cancer has significant correlation, It can significantly increased the risk of lung cancer susceptibility(p=0.005,OR=2.07,95%CI:1.25-3.42)Hierarchical analysis found that CT genotypes can significantly improve the susceptibility to lung cancer in less than 60 year old, male,non-drinking, smoking group. For the rs1948 of CHRNB4 gene exon locus and rs230496 of NF-κB1 intron locus genetic model, there are no associated were observed in the Chinese population lung cancer susceptibility. The rs230496 site p value close to 0.05 over dominant model are analyzed, and the results showed that the site of the AG gene in 60 years old group(p=0.033, OR=0.61,95%CI:0.39-0.96)and group Female(p=0.039, OR=0.56,95%CI:0.32-0.97)plays a protective role.Conclusions:We found a new SNP site which is significantly correlated with lung cancer susceptibility and is located in the intron region of rs4648127 gene of NF-κB1, the CT locus can significantly increase the lung cancer susceptibility, which laid the foundation for us to further explore the function of the gene and lung cancer and vigorous system.
