Objective: To explore the expression of DDX4 in ovarian cancer in vitro and vivo, comparing with CD133 and predict its clinical value.CD133 is the most common tumor stem cell marker. Compare the expression of DDX4 to CD133,and combine the specificity of DDX4,we can guess its potential value on the basis of the research findings of CD133.Methods: 1. The differential transcription level of VASA and CD133 were determined by QRT-PCR in the ovarian cancer cell line SKOV3; 2. The differential expression level of DDX4 and CD133 protein were detected by Western Blot in the ovarian cancer cell line SKOV3; 3. Immunohistochemical method was used in examining DDX4(VASA) expressions in ovarian cancer tissue samples,ovarian borderlian tumors,ovarian benign tumors and normal ovaries.Results: 1. In the ovarian cancer cell line SKOV3,the relative delta delta CT value of VASA m RNA is 0.000133、9.52E-05、0.000103; the relative delta delta CT value of CD133 m RNA is 2.26E-05、3.5E-05、3E-05.The expression of VASA m RNA decreased significantly than CD133 in ovarian cancer cell line.2. The expression of DDX4 protein decreased than CD133 in ovarian cancer cell line: the relative value of protein is the band density of them/ the braid density of GAPDH.The result is that the expression of DDX4 protein is nearly twice than CD133.3. The DDX4 protein was expressed in ovarian cancer tissues, and the distribution was various in different tissue typing: In the tissues of serous cystadenocarcinoma of ovary, cancer cells form like flake or irregular glandular cavity,positive staining in cytoplasm; In the tissues of mucinous cystadenocarcinoma of ovary, tumor cells are large with big and dark stained nucleus, and these cells can secrete mucus. Most of DDX4(VASA) spread around the gland, positive staining in cytoplasm. In the tissues of ovarian clear cell carcinoma, cancer cells are big and round with stained nucleus while the cytoplasm is clear, and DDX4(VASA) mainly spread in the cytoplasm.Conclusion: What is said above, the DDX4(VASA) is likely to become a new ovaian cancer stem cell marker. Over expression of DDX4(VASA) is associated with the development and progression of ovarian carcinoma and there is certain clinical value for understanding the biological behavior,complete early diagnosis of ovarian cancer and improve the prognosis.
