Objective:Exosomes which is about 20-200nm are small vesicles secreted by most cell types in culture.and can mediate diverse biological functions including angiogenesis, cell proliferation, tumor cell invasion and metastasis, immune response, and antigen presentation by the transfer of proteins, mRNAs and non-coding RNAs,such as microRNA, to neighboring or distant cells.Exosomes formintracellularly by inward budding of the multivesicles.Because of the stability and functionality of the exosomal microRNAs,we evaluate and analyze the exosomal miRNA in serum isolated from healthy subjects and patients with coronary artery disease by deep sequeencing.Methods:The patients in the PLA general hospital were selected and divided into stable coronary artery disease group(10)、unstable angina group(10) and acute myocardial infartion group(10)The healthy people(11)were taked into control group. Eeosomes were isolated from the serum of the patients and control group.we observed morphnology of exosomes by transmission electron microscope(TEM) and measure the diameters by nanosight LM10.And we isolate the exosomal miRNA,then we use deep sequeencing to profile miRNA and identify differences among the four groups.Results:The exosomes observed by TEM were Oval and chircle and the diameter were about between 30-200nm,The average diameter were 129nm. From the 41 sequencing libraries, Among the mapped reads, mature miRNAswere 915 in AMI,859in UA,775 in SCAD,822 in Health relatively. The most abundant miRNA found in the serum exosomes were hsa-miR-451b、hsa-miR-451a、hsa-miR-486-1、hsa-miR-486-2、 hsa-miR-486-5p、hsa-miR-486-3p. miR-146b-5p was significantly higher in the patients in acute myocardial infarction group than the health group.Conclusions:serum exosomal miR-146b-5p are promising prognostic biomarkers for acute myocardial infarctionpatients.
