Objective:in vivo, we inject the gingiva-derived mesenchymal stem cells through the caudal vein before and after the radiation respectively in order to observe the effect of prevention and treatment of gingiva-derived mesenchymal stem cells on radiotherapy-induced oral mucositisMethods:1. Isolated and culture for gingiva-derived mesenchymal stem cells in the GFP transgenic mice (C57/BL)2. Building the pathological model of RTOM. The mice are randomly divided into four groups and each group has 32 mice. Experimental group of prevention (30 minutes earlier to inject 1×106 GMSCs before radiation); experimental group of treatment(7 days after radiation to inject the 1×106 GMSCs) and both have its control group(to inject the culture media without GMSCs). Finally observing its effect on the prevention and treatment of RTOMResults:1 The isolation and culture of C57/BL mice’s GMSCs The primary culture cell is isolated and cultured from the C57/BL mice. After culturing GMSCs for 3 days, they were typical fibroblast-like morphology.2 Building the pathological model of RTOM of mice The C57/BL mice are irradiated locally with 25Gy and induced the RTOM. On the 3rd day after radiation, about 51% of mice have the congestion symptoms in tongue and lips. On the 7th day after radiation, there have appeared the symptoms of erosion in lips. On the 10th day after radiation, some mice dead. But in this experiment, have not appeared the mice of class Ⅲ. The Kunming mice are irradiated by the same way and induced the RTOM. These have more serious symptoms than the group of C57/BL mice and have appeared the mice of class III.3 Experiment in vivo3.1 Experimental group of prevention Control group:On the 3rd day after radiation,14 mice have no obvious symptoms,15 mice appeared the symptom of class Ⅰ, three mice death, the occurrence rate of 52%; On the 7th,22 mice appeared the symptom of class Ⅰ,7mice appeared the symptom of class Ⅱ, the occurrence rate of 100%; On the 10th,10 mice appeared the symptom of class Ⅰ,17mice appeared the symptom of class Ⅱ,2 mice death, the occurrence rate of 100%.Experimental group:on the 3rd day after radiation,15 mice have no obvious symptoms,13 mice appeared the symptom of class Ⅰ, the occurrence rate of 46%, four mice death; On the 7th,8 have no obvious symptoms,16 mice appeared the symptom of class Ⅰ,4 mice appeared the symptom of class Ⅱ, the occurrence rate of 71%; On the 10th,6 mice have no obvious symptoms,17 mice appeared the symptom of class I,5 mice appeared the symptom of class Ⅱ,the occurrence rate of 82%. The results, on the 3rd, showed no obvious difference was found between the experimental group and control group (P>0.05); The results, on the 7th,10th, showed obvious difference between the experimental group and control group (P<0.05), which reminder that GMSCs do not have a preventive effect on the early RTOM, but it can reduce the damage of RTOM.3.2 Experimental group of treatment Control group:On the 7th day after radiation(0d of injection), all mice appeared the symptom of RTOM, including 22 mice of symptom of class Ⅰ,6 mice of symptom of class Ⅱ,4 mice death. After 3 days of injection, there are 8 mice for class Ⅰ,18 mice for class Ⅱ,2 mice death; on the 14th day (7th of injection), there are 18 mice for class Ⅱ,8 mice death.Experimental group:On the 7th day after radiation(0d of injection), all mice appeared the symptom of RTOM, including 24 mice of symptom of class Ⅰ,5 mice of symptom of class II, three mice death. After 3 days(3d of injection), there are 6 mice have no obvious symptom,15 mice for class 1,7 mice for class Ⅱ, one mice death,; on the 7th of injection, there are 9 mice have no obvious symptom,11 mice for class Ⅰ,8 mice for class Ⅱ. Through the statistics analysis, the result showed significant difference between experimental group and control group (P<0.05), suggesting the GMSCs have a therapeutic effect on RTOM.Conclusion:Gingiva-derived mesenchymal stem cells do not have the effect of the incidence of RTOM, but it can reduce the damage of RTOM and have the effect of treatment on RTOM...
