| ObjectiveBreast dysplasia is a common clinical adverse modern breast disease, ancient chinese historical records little.Currently,Breast dysplasia prevalence has increased gradually in women and tend to be younger, it affect not only beautiful,but also may affect the health of women and the future of nursing functions.Traditional chinese medicine and modern medicine have made lots of research in breast dysplasia, but there is still no effective product or safe way to treat breast dysplasia.Jiajian Guishen pill is owned by the kidney classical prescription from kidney pill for the treatment of kidney deficiency, fine blood deficiency of a clinical experience prescription,but there is no mechanism related.This subject was based on rethinking the Traditional Chinese Medicine pathogenesis of the breast dysplasia, the animals pattern of mammary gland atrophy treated with Jiajian Guishen pill were detected by applying the theory of modern medicine and experimental methods.The growth of mammary gland was evaluated by the diameter and the height of mammary gland,body weight,estrous cycle,sexual hormones,coefficient of the uterus,spleen and kidney,pathological changes of breast tissue, as well as the expression level of ERaand PR.The objective of this study was to research the effect of Jiajian Guishen pill in curing breast pathology atrophy in rats and to explore its mechanism of action of Jiajian Guishen pill in curing the rats pattern of mammary gland atrophy.MethodsForty-eight adolescent female SD rats were taken as experimental subject and were established the animals pattern of mammary gland atrophy by bilaterally ovariectomized using the classic technique successfully. The rats were divided into 6 groups randomly, which were sham group, model group,positive group and subtraction Jiajian Guishen Pill high-dose, middle-dose and low-dose group, with 8 rats in each group. And oral administration drugs were used in each group for 30d respectively,Detection were taken for the general condition of rats, the change of the estrous cycle, the rats’weight, the organ coefficient of uterus, spleen and adrenal organ coefficient; the serum of E2 and P levels by ELISA,the expression of ERa and PR of the breast tissue by RT-PCR method, and the diameter and height of rat mammary and the pathology changes in rat mammary papilla were observed.Results1.Animal models of breast atrophy:The rats’mammary gland atrophy model was successfully replicated by the removal of ovaries,it showed that rats with hair haggard, irritable, difficult to crawl, such as obvious kidney-yin deficiency signs.The rats’body weight was significantly increased after 14 days (P<0.05),the days of estrus were shorten significantly (P<0.01), the days of diestrus were prolonged significantly (P<0.01),and the level of E2 and P in serum was significantly lower than the sham group (P<0.01).2.The rats’general condition and the change of Estrous cycle and body weight:After administration, compared with the model group,The middle and low dosage of Jiajian Guishen pill could significantly improve the rat hair haggard and alopecia, the rats were easily captured, and the isolation experiments attack points were significantly reduced (P< 0.01), as well as the weight of rats were reduced significantly after administrated 20 days (P<0.01),and the number of rat estrous cycle significantly increased (T<0.01).3.Organ coefficient changes:The organ coefficient of uterus and adrenal and water intake of uterus of model group were decreased compared with the sham operation group(P< 0.05).After drugs intervention, Positive group and the middle-dose,low-dose of Jianjian Guishen pill could increase the rat uterus and adrenal indexes as well as water intake of uterus inordinately (P<0.01,P<0.05),but show no effect on spleen index (P>0.05)4.Changes of serum sex hormones:The serum E2 and P in model rats were significantly lower compared with sham operation group(F<0.05),After administration, Positive group and the middle-dose,low-dose of Jianjian Guishen pill increased the content of serum E2 and P significantly (P<0.01,P< 0.05).5.ERaand PR expression changes:The rat mammary tissue ERa expression in positive group and the middle-dose,low-dose of Jianjian Guishen pill were significantly increased compared with the model group (P<0.01,P<0.05),while the positive group showed no significantly effect on PR expression. The middle-dose,low-dose of Jianjian Guishen pill could increase the expression of PR significantly compared with the model group (P<0.01,P<0.05).6.Mammary gland histopathology changes:The first 2 nipple diameter and height of Positive group and the middle-dose,low-dose of Jianjian Guishen pill increased significantly compared with the model group(P<0.01),In the side of rat mammary tissue structure,Jiajian Guishen pill middle-dose and low-dose group restored remarkably the structure of atrophic mammary gland,and increased the number of mammary glands,enlarged glands bureaucratic and acinar cavity,as well as loosed gland connective tissue,and had no hyperplasia phenomenon, And although positive group could improve the state of rat mammary gland atrophy, but part of the rat mammary tissue fibrosis.Conclusion1.Jiajian Guishen pill could significantly improve the state of the estrous cycle disorders under appropaiate concentration,and ease model rats haggard hair, impatience, irritability and other symptoms of kidney, as well as inhibit the compensatory increase in body weight in ovariectomized rats,greatly improve the obese of ovariectomized rats.2.Jiajian Guishen pill treatment of breast dysplasia were effective than estrogen estradiol valerate preparations in a whole,but in improving body weight and the level of E2,P were considerable with estradiol valerate. And it could effectively inhibit contraction of the uterus and adrenal glands of rats caused by castration.3.Jiajian Guishen pill could round up the breast tissue ERa and PR expression, and improve the structure of breast tissue atrophy in rats, increase significantly rats’mammary gland diameter and height of the nipple and raise the number of mammary glands, relieve the rat mammary gland atrophy,and make up for mammary dysplasia of the breast tissue structure induced by exogenous hormones long time accumulation in vivo.It indicated Jiajian Guishen pill showed a protective effect in clinical treatment on mammary gland’s structure.On the above studies, Jiajian Guishen pill could promote female adolescent rats’ mammary gland development, through the way of adjusting Endocrine system、Sex hormone level and sex hormone receptor’s expression and The morphological of rats’ mammary gland. |
