Objective: To study the chemical components of Paeonia suffruticosa Andr.(named tree peony).The paper explore the formation mechanism of Feng Dan through the fingerprint,the Q-TOF spectrogram,volatile oil,inorganic elements analysis and of Cortex Moutan from different origins.Methods: The UPLC method was used to creat the fingerprint method of Cortex Moutan which was the reflux extraction of Cortex Moutan. Based on the fingerprint method,the Q-TOF technology was used to identify its chemical components with improving the conditions of chromatography and mass spectrometry.The volatile components Cortex Moutan of were extracted by the ultrasonic,the quantitative and qualitative analysis was given by GC-MS technology.The microwave digestion method was used to process the raw material,ICP-MS and ICP-OES technology combined with thesemi quantitative method was used in the elemental qualitative and quantitative analysis of Cortex Moutan.We adopt the similarity calculation software, SPSS, simca-p and other data processing software for the analysis of the above results, and analyse and identify the possible marker which reveals the differences in their chemical composition of Cortex Moutan.Combined with ecological factors(temperature, rainfall, soil analysis and so on), we suggest which ecological factor may influence the genuineness formation of Feng Dan.Results: Through the study of UPLC fingerprint of Cortex Moutan, we got a fine analysis method which can be separated well,and the baseline was stable, quick and effective.It can evaluate Cortex Moutan on the whole. The application of Q-TOF in Cortex Moutan were identified by chemical analysis, a total of conjecture 37 ion peaks of chemical structure, can be divided into benzoic acid, paeonol, glucose,gallic acid class peony glycosides, flavonoids and other five kinds of compounds.Multivariate statistical analysis obtained from different origin differences in quality and quantity of material exists,the potential 5 kind of marker was possible Paeonolide and so on through analysis and identification.The GC-MS study on volatile components of Cortex Moutan, identified a total of 41 chemical constituents from 5 origins, which was mainly for benzoic acid, gallic acid and paeonol, long chain fatty acids, multivariate statistical analysis described their chemical components’ differences in the content and kind.The marker was possible 2- amino ethanol and so on. Combined ICP-MS with ICP-OES,the relationship of the herbs’ quality and inorganic elements was analyzed.There were significant differences in the Feng Dan(from Tongling) and other areas.The results was found that Cu, As, Cd, Pb, S, Fe and other elements were higher than those from other origins,multivariate statistical analysis results display Li, Zn, P, Fe, K and other elements may be the main factors different from other origins.Conclusion: It established the combination technology analysis method(included UPLC, Q-TOF,GC-MS, ICP-MS,ICP-OES etc.) that analyze the chemical components of Cortex Moutan from different origins.Combined with multivariate statistical analysis,we found that there were different degrees of difference in content and species in the chemical components compared with the famous region drug.Climate and soil factor may be the main factor in the formation of its genuineness based on the study of chemical composition and ecological factors, the ecological factors of Tongling are more suitable to cultivate Cortex Moutan and accumulate active ingredient.The comparative study of Cortex Moutan provides the experimental basis for the material basis of elaborate Feng Dan medicine and genuineness of formation mechanism, but also lays the foundation for the quality evaluation method and quality standard of Feng Dan.
