Objective: To ex Plore the intervention effect of Pingchuanning mechanism of cold asthma rats lung tissue in ERK signal transduction Pathway in the treatment of cold asthma. Through the ERK signal transduction Pathway observed in asthma rat s induced by factor PDGF、RAS、MEK1、ERK1、C-FOS、P-ERK1 and other related Protein, investigate dex Pression and asthma related indexes of airway rem odeling of matrix metallo Proteinase MMP-9 and matrix metallo Proteinase inhibit or TIMP-1 dry Ning, discuss the effect mechanism of Pingchuan ning inhibit airw ay remodeling cold asthma rats.Methods: Selected from the ex Perimental animal center of Medical University Of Anhui SPF class male SD rats, weight at 220±20g, 140, were randomly divided in to normal grou P、model grou P、Ping chuanning high dose grou P、Ping chuanning middle dose grou P、Ping chuanning low dose grou P、dexamethasone grou P and G uilong Kechuanning grou P,a total of 7 grou Ps, each grou P of 20 only. Using the 10% egg Protein solution and cold stimulation combined with sensitized rats,cold ast hma model to co Py the success. In a successful after 21 days by intragastric admini stration, while the normal grou P and model grou P were given normal saline every day.Remove the lung tissue after 4 weeks, the left lung with formalin for HE staini ng to observe the ex Pression of Preservation, the lung tissue Pathological changes a nd immunohistochemical method to detect MMP-9 and TIMP-1 in lung tissue; right lung into-80 C refrigerator Preservation, used for Western blotting(Western Blo t) ERK signal Pathway induced lung tissue factor PDGF mouse detection, signal trans duction Pathway of PDGF、RAS、MEK1、ERK1、C-FOS、P-ERK1 and other rel ated Protein ex Pression.Results: Through HE staining can be found, com Pared with the normal grou P, model grou Phad obvious airway remodeling Phenomenon; com Pared with the model g rou P, the treatment grou Ps were Pro Perly alleviate airway remodeling. By immunohi stochemical method was com Pared with the normal grou P, the ex Pression of MM P-9、TIMP-1 has the higher, the difference was statistically significant(P<0.01, P<0.05); and com Pared with the model grou P, Pingchuanning thetreatment grou P MM P-9、TIMP-1 ex Pression was decreased Pro Perly(P<0.01,P<0.05) es Pecially in hi gh dose grou P decreased the Pingchuanning obviously, the best effect. Ex Pression of by detecting the ERK signal transduction Pathway of WB by PDGF、RAS、MEK1、ERK1、C-FOS、P-ERK1 and other related Proteins com Pared with the normal grou P, ex Pressed in PDGF、RAS、MEK1、ERK1、C-FOS、P-ERK1 and other related Protein in asthmatic model increased(P<0.01,P<0.05); com P ared with model grou P, small dose grou P in Pingchuanning in the down-regulation of P-ERK1、ERK1 ex Pression is not obvious, is ex Pressed in the grou Ps PDGF、RAS、MEK1、ERK1、C-FOS、P-ERK1 and other related Proteins(P<0.01,P<0.05) a PPro Priate decline.Conclusion:Asthma rather by inducing factor PDGF、RAS、MEK1、ERK1、CFOS、P-ERK1 and other related Proteins and the ex Pression of matrix metallo Prot einase MMP-9 and matrix metallo Proteinase inhibitor TIMP-1 to alleviate the airwa y inflammation of asthmatic rats regulate ERK signaling Pathway, and can effec tively reverse the airway remodeling and treatment of asthma.
