ObjectiveStudy the effect of berberine combined with cinnamaldehyde to REM sleep deprivation rats blood and brain tissue distribution,which can prove prescription combination rule, and develop new insomnia medicine, provide ideas and methods.MethodsThe 40 rats were randomly divided into four groups:SD rats were divided into normal control group, model group, berberine with cinnamic aldehyde group (at a dose of 0.65g/kg X d), berberine group (at a dose of 0.5g/kg X d), the use of sleep deprivation model of rat small platform Water Environment Act, liquid chromatography-mass spectrometry (LC-MS) was used to detect the distribution of berberine in blood and brain tissueResultsCompared with normal control group, model group’s sleep deprivation was successful, in which the rats wholely were in hyposomnia. Berberine cinnamic aldehyde group (dose:5g/kg×d), the berberine distribution level of these rats is higher than berberine group (without cinnamic aldehyde) (P<0.05). After gavaging 1.5 hours, the berberine distribution level of Berberine cinnamic aldehyde group is higher than berberine group (without cinnamic aldehyde) (P<0.05)Conelusionberberine combined with cinnamic aldehyde, sedative and hypnotic effects enhanced berberine in blood and brain tissue metabolism and an increased content distribution, and its mechanism berberine may reduce the effect by cinnamic aldehyde metabolic rate in the blood and increase the blood-brain barrier permeability related.
