ObjectiveTo observe the change of TNF alpha, cTnI and h-FABP in CLP septic mice’ serum, and study the correlation between the expression of inflammatory cytokine and the level of myocardial damage. Base on upon results, to study the effect of the Resveratrol by different dosages for myocardial damage on CLP septic mice.Methods1. forty-eight male C57BL/6 mice was mainly randomly divided into four group:Normal group, Sham-operation(short for Sham) group, CLP group and Resveratrol (short for Res) group. Res group was divided into three different groups by different dosages:Res20 group(20mg/kg), Res40 group(40mg/kg), Res80 group(80mg/kg).2.The mice of Normal group, Sham group and CLP group were received peritoneal injection with 0.4ml distilled water 2h after the operation. The mice of Res group were received peritoneal injection with 0.4ml Resveratrol by different dosages 2h after the operation. Blood samples of each group were obtained from Orbital venous plexus at 6,12 and 24 hours after operation. The blood serum was saved after centrifugation.3. The levels of TNF alpha, cTnI and h-FABP were determined by ELISA.4. All data were conducted by SPSS 17.0 Statistic Package for Social Science. All data were expressed by the mean and standard deviation. The data between two different group were compared by one-way analysis of variance, the difference was statistically significant if P<0.05. Two variables related analysis by linear correlation analysis. The data of Heterogeneity of variance was analyzed by Dunnett t-test. The data of Homogeneity of variance was analyzed by Fisher’s least significant difference t-test. The relevant degree was showed by Pearson Product-moment correlation coefficient.Results1. The mice of CLP group appeared to the changes of normal state, while the symptom of Resveratrol group is less severe, and the mortality of Resveratrol group was lower than CLP group.2. The correlation between the level of TNF alpha and those two myocardial biomedical markers is positive by the analysis of all data.3. After CLP 6 hours, compared with the normal group and sham operation group, the level of TNF alpha, cTnI and h-FABP were significantly increased(P<0.01). Moreover, the level of all the them were increased along with the time.4. Compared with CLP group, the level of TNF alpha, cTnI and h-FABP of Resveratrol group were significantly decreased after CLP 6,12 and 24 hours(P<0.01). However, all of them were higher than the normal group and the sham operation group.6. By the observation of the change of TNF alpha, cTnI and h-FABP at different timing, it shows that the remarkable difference was not found among the three different Resveratrol groups.Conclusion1. The correlation between the level of the expression of inflammatory cytokine and the level of myocardial damage is positive.2. Based on the result of the test, Resveratrol might lighten the myocardial damage by reduce the expression of inflammatory cytokine.
