Licorice is one of the most widely used traditional Chinese herbal medicines, and it rich in proteins. The thesis focused on the proteins of Licorice, and the contents involved isolation, purification and properties characterization of native proteins derived from Licorice, self-assembly conditions optimization of licorice protein, the stability and drug-loaded of nanoparticles experiment. It is discussed the self-assembly behavior of protein derived from traditional Chinese herbal medicines in a new perspective.A new protein GP have been separated and isolated from licorice by High Q anionic exchange and POROS(?)R1 hydrophobic interaction chromatography, buffer extraction and ethanol sedimentation before. Just a single band appeared in SDS-PAGE, which molecular weight is 28.0 kDa. IFE display its pI is 4.5. Result of N-terminal sequencing by Edman degradation is NPDGLIACYCGQYCW, there returns no similar results when search in NCBI.Nanoparticals GPNs formed by holding 60 min in 100℃,concentration of GP is 0.9 mg/mL, which Z-ave is 74.09±0.69 nm, and its Zeta potential absolute value is 24.3±1.69 mV. Morphological characteristics of GPNs observed by SEM, and found that it was spherical approximately. The result of assembly yield displayed that 87.9% GP assembled into GPNs.The result of molecular weight showed that 22 GP moleculars assembled into GPNs approximately in perfect state by MALLS/GPC.GPNs can storage in normal temperature preservation was just appropriate for whose need storage in short-term by stability test of GPNs. Freeze will help to save for a long time.Both GP and GPNs were not restrained cell increment, which showed low cytotoxicity. GPNs can combine with cell surface specifically, GP can not.The GPNs are able capsulize 51.2% Astragaloside IV into it, then its volume increased.This is the first time using protein derived from licorice assembled into GPNs, it is expected to be a novel drug carrier material. Meaningwhile, it offers a new approach to the modernization of traditional Chinese medicine.
