Background:Metabolic syndrome was a constellation of modifiable vascular risk factors, The main components of the metabolic syndrome was obesity, diabetes mellitus, with high levels of blood triglycerides (TG) and low levels of HDL for the characteristics of dyslipidemia, hypertension. With China’s rapid economic development, Coming daily home Structures of life and lifestyle have changed obviously, the prevalence of the metabolic syndrome has increased by years.It has bringed such a stressful burden on the home as well as the whole society. The growing emphasis on the Early diagnosies and treatment of the metabolic syndrome was essential.The clinical and epidemiological showed that the metabolic syndrome was also a complicated pathological process involves many factors, many genes and many stages.It is muitifactorial, so far it is still unclear. If we can study metabolic syndrome from gene level, we can identify and primary prevention of the metabolic syndrome. Therefore this research was investigate the association of CD36 rs10499859 Single Nucleotide Polymorphism with metabolic syndrome.It was practical significant.Research purposes:The CD36 gene polymorphism were determined by the polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) technique.The purpose of this study was to know the distribution characteristics of CD36 rs 10499859 Single Nucleotide Polymorphism, to explore the relationship between CD36 gene rs10499859 polymorphism and metabolic syndrome in the GuangXi Province population. We investigated the risk factors of metabolic syndrome into the genetics.Methods:Totally consecutive 119 cases in the patient group of metabolic syndrome were collected at the First affiliated hospital of Guangxi medical university during December 2012 to June 2014.69 cases were male, and 50 cases were female.89 cases in the control group without Diabetes and Hypertension were collected at the Physical examination center of the First affiliated hospital of Guangxi medical university. The level of height, weight, fasting blood glucose, 2-hours postprandial blood sugar total cholesterol(TC), triglyeeride(TG), high density lipoprotein (HDL-C) and low density lipoprotein(LDL-C) were measured in all the patients with 12 hours fasting overnight. The level of the indicators were determinated in the first affiliated hospital of Guangxi medical university test center. Body mass index (BMI) was calculated. The patients group and the control group were Extracted the venous blood 2ml, and separated into blood cells and plasma and isolated the white blood cells immediately, stored at-20 ℃ for extraction of genomic DNA. We used the phenol-chloroform method to extract DNA, and the extracted DNA of CD36rs 10499859 genotypes were detected by PCR-RFLP method. The results were analysed by the SPSS 16.0 statistical software.Result:1.The distinction and frequency of gene between the control group and patient group:The genotype frequency of AA,AG and GG did not show a significant difference(P>0.05). It did not show a significant difference in the allelic (A and G) frequency of CD36 rs 10499859 gene (P>0.05).2. Laboratory testing in the three genotype of CD36 gene rs10499859 compared in the two groups:Three genotypes (AA,AG and GG) were compared in pairs in the patient group,the level of HDL-C in the GG genotype carriers was significant higher than the AA genotype carriers(P<0.05);the rest of the indicators was no statistically significant difference between the genotypes (P>0.05).3.The logistic regression analysis showed the risk factors of metabolic syndrome, based on whether the metabolic syndrome as the dependent variable, whlile the age, systolic blood pressure,2 h postprandial blood glucose, fasting glucose, BMI, triglyceride, HDL-C, LDL-C and rs10499859 genotype as the independent variable. The results showed that age into the regression equation, The value of OR was1.371.(P<0.05), the rest of the variables are not into the equation.Conclusion:1.The Single nucleotide polymorphisms of CD36 rs10499859 had no direct relation with metabolic syndrome in the GuangXi Province population.2.The Single nucleotide polymorphisms of CD36 rs10499859 was associated with the level of HDL-C, mybe it was a protective gene for metabolic syndrome in the GuangXi Province population.3.The risk factor of metabolic syndrome was the age, if the age increased 1 year, that would increase 37.1% risk for metabolic syndrome.
