The Discussion About The Aging Mechanism Of Zuo Guiwan And You Guiwan Reversed POF Induced By Chemotherapy Based On The E₂、P Level And Its Receptor Correlation

Objective:In POF rats model induced by chemotherapy,in different period of chemotherapy and in different period of sexual cycle,using zuogui wan to nifying kidney-yin or using yougui wan tonifying kidney-yang, observe the effect of the E2、P level of rats,and the effect of the reaction of rat uterine ERα, PR on E2, P. Analyse the relevance between E2, P levels and ERα, PR, to investigate the mechanism of zuogui wan and yougui wan resistance of ch emotherapy induced POF in different period and zuogui wan and yougui wan re storation of ovarian reserve during the menstrual cycle in several different periods and the protection, to reveal the scientific connotation of " kidney controlling reproduction", to clarify the effectiveness of "prevent disease before" and "TCM cycle therapy". Methods:Choose 220 female SD rats which is 12 weeks old, monitor each female rat vaginal exfoliated cells, establish sexual cycle archives, and choose the rat which has regular sexual cycle. The experiment was divided into three parts, there are six groups in each part: blank group; model group; estrogen and progesterone group;zuogui wan grou p;yougui wan group;zuogui wan and yougui wan group. The different of Molding and drug intervention of 3 parts of experiment is, part 1(early intervention): CTX Molding and drug intervention at the same time, for 15 days.Part 2(sync hronization and late intervention): Molding and drug intervention at the same time, for 15 days,then drug intervention for 20 days.Part 3(late intervention): CTX Molding for 15 days, Divided successful model rats into five groups, drug Intervention for 20 days, After the last administration which in 1-5 days in proestrus or fifth days did not occur during the estrous cycle of rats were put to death,draw 5ml blood from the abdominal aorta, excision rat uterus checking weigh and saving,and detection the experimental index. Detected the serum E2, P levels of the rats in each group by radioimmunoassay; detect the relative expression level of the ERαm RNA of the rats uterus by the realtime fluorescence quantitative PCR. Detect the expression of PR protein by Immun ohistochemical method. Results:1. All kinds of index of the rats model of chemotherapy in the three part of the experiment shows the changing of prema ture ovarian failure, the E2、P level、the relative expression level of the ERαm RNA of the rats uterus and the expression of PR protein of each model group are Significantly lower than the value of blank group,it is quite si gnificant(P<0.01).2. Early intervention and late intervention:the E2、P level of model group significantly higher than estrogen and progesterone group, it is quite significant(P<0.01);the E2,P level of zuogui wan and yougui wan group was raised, it is significant(P<0.05). The relative expression level of the ERαm RNA of the rats uterus and the expression of PR protein of zuogui wan and yougui wan group and estrogen and progesterone group significantly higher than model group(P<0.05). The effect of zuogui wan group and yougui wan group was not obvious.3. synchroniza tion and late intervention: the E2、P level、the relative expression level of the ERαm RNA of the rats uterus and the expression of PR protein of zuogui wan and yougui wan group and estrogen and progesterone group significan tly higher than model group, it is quite significant(P<0.01). The effect of zuogui wan group was more obvious than the effect of yougui wan group.4.The E2、P level、the relative expression level of the ERαm RNA of the rats uterus and the expression of PR protein in part 2 significantly higher than the value in part 1 and part 3. Conclusion:Zuogui wan and yougui wan can improve the general condition of the POF rats; The E2、P level of POF rats falling can decrease the ERα、PR level;Zuogui wan and yougui wan can increase the E2、P level of POF rats, so them can increase the level of the ERαm RNA of the rats uterus and the expression of PR protein; Sequential therapy of TCM-yin and Yang is more effective than only use zuogui wan or only use yougui wan; Synchronization and late intervention has obvious advantages to only early intervention or only late intervention in the trea tment of chemotherapy induced POF rat with zuogui wan and yougui wan.
