The Effect Of Juanbi Capsule On The Expressions Of TLR4 And NF-κBp56 In Guinea Pig Primary Osteoarthritis Synovial

Objective:To observe the effect Juanbi capsule on animal guinea OA synovium TLR4, NF-κBp56 and TNF-α expression.Method: The 40 one month-old female guinea pigs taken eight straight drawn as normal,, and the remaining 32 as a model group feeding to 3 months, establish OA guinea pig model, then month-old guinea pigs were randomly divided into model group(3 months), model group(5 months), glucosamine hydrochloride capsules group and Juanbi capsule group, n=8 average. Model group(3 months) comes directly from the model group(5 months old) give water orally, glucosamine hydrochloride capsules were given glucosamine hydrochloride capsules orally, Juanbi Juanbi capsule capsules were given orally, the above three groups ig two months. Two months after the general observation of experimental animals, histological observation. Using a modified Mankin’s scoring methods quantitative analysis of cartilage damage in different degrees; immunohistochemical method for determining bone TLR4 expression, NF-κBp56 and TNF-α protein into the synovium and cartilage under the last application software Graph Pad Prism5.0 The results were statistically analyzed.Results:(1) The results of each group Mankin’s score:as shown in Figure A, compared with the normal group, model 3-month-old group was significantly higher,with statistical significance,p<0.05;and model 5-month-old group, the amino sugar group and Juanbi group Mankin’s score was significantly lower, p<0.05;Juanbi group Compared with Glucosamine group,p<0.05;(2) immunohistochemical results TLR4 comparison groups: the model group than in the model 5 months 3 months TLR4 expression was significantly higher,p<0.0001;give Juanbi capsule treatment, and model 5-month-old group,TLR4 significant reduction, p<0.0001;Juanbi group and Glucosamine group, TLR4 expression lower Juanbi group,p=0.0112;(3)NF-κBp56 immunohistochemical results of each group: Model 5-month-old group than 3 months old model NF-κBp56 expression wassignificantly higher,p=0.001; after giving Juanbi capsule treatment group compared with 5 months old model, NF-κBp56 significant reduction,p<0.0001; Juanbi group Compared with Glucosamine group,Juanbi lower expression of NF-κBp56 group,p=0.0188;(4) immunohistochemical results of each group TNF-α comparison: the model group 5 months 3 months TNF-α expression was significantly compared with the model increase, p<0.0001; after giving Juanbi capsule treatment group compared with 5 months old model,TNF-α decreased significantly,p<0.0001;after administration of glucosamine hydrochloride treatment, the expression of TNF-α were significantly lower than model 5 months group, p<0.0001;compared Juanbi group TNF-α expression Juanbi group and lower Glucosamine group, p=0.0078.Conclusions:(1) Juanbi capsules and glucosamine hydrochloride capsules can significantly reduce the guinea early OA animal model of joint Mankin’s score, and inhibit the expression of synovium TLR4, NF-κBp56 and TNF-α, but Juanbi capsule effect is more significant.(2) Juanbi capsules can reduce the degeneration of female guinea pig animal model of OA cartilage and inhibit synovial inflammation.
