Objective:By observing dynamic change of the early fatty liver rats serum leptin and expression of liver uncoupling protein 2, To explore the relationship between phlegm and blood stasis with leptin and uncoupling protein 2 in the development of early fatty liver. Methods:1, Model preparation: Give numbers to 140 SD rats, randomly divided into 4 groups. Normal group(50) give the ordinary feed, model group(50) and simvastatin group(20) and modified Erchen decoction group(20) give the high fat feed.2, Grouping dosing: Gavage normal group and model group to saline began from 8 weekend, gavage simvastatin group and modified Erchen decoction group respectively to simvastatin and modified Erchen decoction untill 12 th weekend.3, In 4th, 6th, 8th, 10 th, 12 th weekends, dynamic observe each group rats weight, liver weight and liver index, serum liver function(ALT, AST), blood lipid(serum TC, TG, HDL- C, LDL- C), liver fat(liver TC, TG) and fasting blood-glucose(FBG).Liver tissue paraffin section with HE staining, dynamic observe the hepatic steatosis.Using Enzyme linked immunosorbent assay(ELISA) testing each groups rats serum leptin(Leptin) content, using rt-pcr(realtime PCR) detection each groups rats liver uncoupling protein 2(UCP2) m RNA expression.Results:1, General situation: compared with normal group, model group rats weight, liver weight and liver index increase in 6th, 8th, 10 th, 12 th weekends, the difference was statistically significant(P < 0.05).2, Pathology: The model group rats pathologic shown form 4th weekend liver cell volume increase in part, certain sizes of lipid droplets cavity can be seen in the cytoplasm of liver cells;In 6th weekend hepatic cord disorders and cell swelling, more liver cell cytoplasm appear different size of round fat drops and cavitation, part of the fusion of fat cavitation nuclei to one side;Structure of hepatic lobule is not clear in 8th weekend, hepatic cord radial alignment is not obvious, liver cell volume increase, liver cell sizes can be seen in the cytoplasm of lipid droplets cavitation, fusion of lipid drops after cavitation nuclei can be pushed to the cell membrane, then the perspective of per unit area of more than one-third of the hepatocyte fatty degeneration can be seen under microscope;Disorganized in 10 th weekend liver and liver cell swelling, more than two-thirds of the liver cells in the cytoplasm of different sizes round fat drops of cavitation, part of the cavity after fusion to the nucleus into the cell membrane;Disorganized in 12 th weekends liver and liver cell swelling, cytoplasm loose net-like, translucent(cytoplasm loose), severe cases almost completely transparent cytoplasm(balloon sample).3, Serum biochemical: Compared with normal group, model group serum ALT, AST increased in 12 th weekend, the difference was statistically significant(P < 0.05);Serum TC, TG, LDL- C in 4th ~ 12 th weeks increased progressive, the difference was statistically significant(P < 0.05);Serum hdl-c leel reduced gradually in 8th, 10 th, 12 th weekends,the difference was statistically significant(P < 0.05);Compared with model group, serum TC, TG of modified Erchen decoction group is reduced in 10 th, 12 th weekend, the difference was statistically significant(P < 0.05); Serum hdl-c leel rised in 12 th weekend, the difference was statistically significant(P < 0.05).4, Liver fat:Compared with normal group, liver fat TC, TG of model group rised in 4th~12th weekends, the difference was statistically significant(P < 0.05);Compared with model group, modified Erchen decoction group of rat liver TC, TG reduced in 10 th ~ 12 th weekends, the difference was statistically significant(P < 0.05).5, Serum leptin: Compared with normal group, model group rats serum leptin increased in 8th, 10 th, 12 th weekends,the difference was statistically significant(P < 0.05);Compared with model group, modified Erchen decoction group reduced in 10 th, 12 th weekends, the difference was statistically significant(P < 0.05).6, Liver uncoupling protein 2: Compared with normal group, model group rats liver uncoupling protein 2(UCP2) m RNA expression rised in 4th, 6th, 8th, 10 th, 12 th weekends, the difference was statistically significant(P < 0.05);Compared with model group, modified Erchen decoction group reduced in 10 th, 12 th weekends, the difference was statistically significant(P < 0.05). Conclusion:1, By high fat feed rats, typical liver fatty change appeared in 8th weekend and prompt early fatty liver rat model copyed success.2, Early fatty liver rats serum leptin level rising in the process of occurrence and development of early fatty liver, and is closely related to the changes of blood lipid, indicate that serum leptin participated its lipid metabolic disorder and early occurrence of fatty liver.3, Early fatty liver rat liver UCP2 m RNA expression level rising in the process of early fatty liver development in rats, and is associated with blood glucose, suggests that UCP2 involved in glucose metabolism and the occurrence of early fatty liver.4, Application of modified Erchen decoction disproof after intervention therapy, model group rats lipid can been improved, the degree of steatosis reduced.Flavored after modified Erchen decoction intervention,model group rats serum leptin expression level decreased obviously, the liver UCP2 m RNA expression reduced, show that in the early stages of the development of fatty liver, serum leptin and UCP2 m RNA may be associated with phlegm and blood stasis.
