[Objective]To evaluate whether the ET instrument is accurate, in quantitative determination of glucose and lactate levels in whole blood samples from clinical patients. A comparative analysis is performed between the ET instrument and an established in vitro technique at Navy General Hospital of PLA, for quantitative glucose and lactate determination in whole blood,to investigate factors affecting analytical results. Finally, conclusions are drawn on whether the enzyme thermistor could be used for on-line and real-time analysis in clinical environment.[Methods]After obtaining the understanding of patients and their family members, an off-line comparative evaluation of patients’peripheral vein blood between ET instrument (the LUND University, Sweden) and LifeScan OneTouch(?) was performed,to analyze glucose and lactate levels in one hundred and eight blood samples at Neurosurgery Department of Navy General Hospital.[Results]1. The linear range of detection was 0.5-30.0 mmol/L and 0.25-12.00 mmol/L for glucose and lactate. The linear regression values were 18.4 and 9.5 for glucose and lactate, respectively.2. Both glucose and lactate for sample concentrations were in the interval of 4.0-6.2mmol/L and 0.5-1.3mmol/L, respectively.3. Comparative analysis between the ET instrument and LifeScan OneTouch (?) showed good correlation.[Conclusions]The principle of the ET instrument is different from other blood analyzers in clinical use.There is good correlation between ET and LifeScan OneTouch(?) by comparing analysis. Various factors affecting the analytical quality were investigated including sampling condition, patient disease and operational setting. Patients with diabetes and heart problems have elevated blood concentrations about glucose and lactate, comparing with our experiment consequence. Thus, the ET instrument accuracy for blood concentrations still needs to be investigated. As the advance of miniaturization, this instrument will be able to perform on-line and real-time blood analysis in clinical environment. Furthermore, its applications also involve food production, environmental monitoring and war industry etc.
