Related Factors Analysis between Plasma Levels of Omentin-1 and Colorectal CancerObjective :To investigate the relationship between plasma adipocytokine omentin-1 levels and colorectal cancer(CRC).Method : This study was conducted among 42 patients with pathologically diagnosed CRC after operation, as the case group and 42 healthy subjects who underwent total colonoscopy because of a voluntary health check-up as the control group. The above two group was matched between age, sex. After participants has fasted overnight on the same day of the endoscopic check their blood samples were collected. Fasting plasma omentin-1 was measured using enzyme-linked immunosorbent assay(ELISA) methods. The following parameters were measured and compared within each group and between groups by a statistical soft system SPSS 20.0. A two-tailed t-test was used to compare the values across groups..Pearson correlation analysis and multivariate linear regression analysis method, analysis of serum Omentin- 1 and the relationship between other variables.Inspection level p = 0.05.Results : In which the patients and healthy people had no statistically significant differences in gender, age, systolic blood pressure(SBP), diastolic blood pressure(DBP), waist-hip ratio(WHR),body-mass index(BMI). serum triacylglycerol(TG), high density lipoprotein cholesterol(HDL-C), and also fasting blood glucose(FBG). The total cholesterol(TC) levels in healthy people was4.83±1.08 mmol/L and totalcholesterol levels were decreased in CRC patients with the value at 3.27±1.14 mmol/L(p<0.05). Compared with the controls, patients with CRC had significantly higher circulating omentin-1(71.54±31.80 vs 31.22±19.87 ng/m L, p < 0.001) low density lipoprotein cholesterol(LDL-C)(2.64±0.63 vs 1.59±0.82 mmol/L, p < 0.05) FINS(fasting insulin)(9.44±6.11 vs 7.03±3.49 m U/L, p < 0.05) levels and HOMA-IR(homeostasis model assessment)( 0.94±0.43 vs0.61±0.33, p < 0.05).Pearson correlation analysis showed that plasma omentin-1 was positively correlated with high density lipoprotein cholosterol(HDL-C)(r=0.57,P<0.01),and negatively correlated with serum triglyceride(TG), fasting insulin(FINS), waist hip ratio(WHR), model insu Lin resistance index(HOMA-IR)(r=-0.668、-0.493、-0.395、-0.371,respectively, P<0.05~0.01).Mu Ltiple stepwise regression analysis showed that TG, HDL-C, WHR were the influencing factors of plasma omentin-1 in CRC patients.Conclusion : The plasma omentin-1 level may correlate with the development of CRC.Effect of Omentin-1 on Biological Behavior in Colon Cancer SW480 CellsObjective: To observe the effects of the omentin-1 with different concentrations on the proliferation and apoptosis of colon cancer SW480 cells in high glucose environment and discuss the effects of signal passage Akt in the process of omentin-1-induced apoptosis in colon cancer cells apoptosis.Methods: The omentin-1 with different concentrations(0 μg/m L, 1μg/m L, 5μg/m L) were used for intervening the colon cancer SW480 cell lines cultured in the high glucose medium(DEME culture medium with glucose content of 4.5g/L) for 24 h and 48 h. The experiment was divided into high glucose +0 μg/m L omentin-1 group(omentin0 group, without omentin-1), high glucose +1μg/m L omentin-1 group(omentin1group), high glucose +5μg/m L omentin-1 group(omentin 2 group), Colon cancer SW480 cell lines cultured in the low glucose medium(DEME culture medium with glucose content of 1.0g/L) was collected as control group(NC group). The cell proliferation activity was tested by MTT methodl. And we added 4μg/m L cisplatin in each group. apoptosis rate was tested by flow cytometer. The protein expression levels of p-AKt and AKt were determined by Western blot.,Results:(1) After 24 h of omentin-1 intervention to SW480 cells, the cell optical density value(OD value) in omentin2 group were increased significantly compared with omentin 0 group and omentin 1group(P<0.05). After 48 h the OD values in omentin 1group, omentin 2 group were increased significantly compared with omentin 0 group(P<0.05). And the OD values among omentin 1 group and omentin 2 group were significantly gradually increased(P<0.05).(2) We detected cell migration through scratches experimental,cell migration resu Lts showed that the experimental group was significantly promoted in 24 hours, more apparent in 48 hours.(3) Colony experimental results show that compared with the control group, the experimental group regardless of clonogenic cell number or size to be greater than in the control group(4) After omentin-1 and DDP intervention SW480 cells, the resu Lts showed that the omentin 1 group and omentin 2 group withered the number of apoptotic cells was significantly lower than that of the control group(P<0.05).(5) The results of Western-blot showed that the protein expression levels of active p-Akt inomentin1 group and omentin2 group were much higher(P<0.01) when compared with omentin0 group, meanwhile compared with omentin1 group, the protein expression levels of active p-Akt of omentin2 group were significantly increased(P<0.05).Conclusion(1) In the high glucose environment, omentin-1 can induce the proliferation of colon cancer SW480 cells, the effect showed the dependent relationship with concentration and time,and omentin-1 can restraint the apoptosis, the effect showed the dependent relationship with concentration.(2) In the high glucose environment, the promoted apoptotic effects of omentin-1 may be associated with adiponectin by activating Akt pathway.(3) Omentin-1 may promote the proliferation, migration and apoptosis of colon cancer cells,...
