Objective: To investigate the prophylactic use by observing rosuvastatin on vascular endothelial growth factor in rats with hire-hypertension of acute coronary syndrome, asymmetric dimethyl arginine influence, improve endothelial function explore possible mechanism for the clinical application rosuvastatin for reliable experimental basis.Method: In this study, male rats were randomly divided into three groups: normal SD rats in the control group(NC group), H-hypertension model group without rosuvastatin(ACS group), H-hypertensive rats were given rosuvastatin intervention group(RCS group),each group was eight rats. H-hypertensive model using spontaneously hypertensive rats fed rat chow add 2% methionine.The rosuvastatin treatment group received 10 mg / kg / d orally, the normal control group and model group H-hypertensive rats fed such as the capacity of distilled water for 4 weeks. During the experiment rats were fed ad libitum water. After four weeks, measured angular vein blood serum homocysteine, total cholesterol, triglycerides, high-density lipoprotein cholesterol, low-density lipoprotein cholesterol. Isoproterenol(30 mg/kg) was injected subcutaneously for one day to induce myocardial infarction. Enzyme-linked immunosorbent(Enzyme-Linked Immunosorbent Assay, ELISA) method to measure serum vascular endothelial growth factor(vascular endothelial growth factor, VEGF) and asymmetric dimethyl arginine(Asymmetrical dimethylarginine, ADMA) protein content change; immunohistochemical expression was observed in the rat vascular VEGF each group. Measurement data are used(s), treatment with SPSS19.0 statistical package, using F-test analysis of three groups according to the different requirements of the above data, the mean variance between pairwise comparisons using LSD-t test when Sarkozy, two sets of data using an independent T-test, with P<0.05 indicate a statistically significant difference.Results:1 Before modeling the blood pressure in spontaneously hypertensive rats in each group did not differ(P<0.05), but significantly higher than normal SD rats. RCS group, blood pressure compared with before treatment there was a significant difference(P<0.05), RCS group comparison between blood pressure and ACS group, the difference was statistically significant(P<0.05).2 Homocysteine, total cholesterol, triglycerides, high-density lipoprotein cholesterol, low-density lipoprotein cholesterol levels change: After four weeks of treatment in rats fed, RCS group and ACS group blood homocysteine content higher than NC group. Confirming H hypertensive rats modeling success,the rats comparison between lipid levels, the difference was not statistically significant(P>0.05).3 Determination of serum levels of VEGF, ACS group, serum levels of VEGF RCS group compared with before treatment There was a significant difference(P<0.05),the VEGF serum content of RCS group was significantly higher than ACS group and NC group, the difference was statistically significant(P<0.05).4 Determination of serum levels of ADMA, ACS group, serum levels of ADMA RCS group compared with before treatment There was a significant difference(P<0.05),the ADMA serum content of RCS group was significantly higher than ACS group and NC group, the difference was statistically significant(P<0.05).5 Immunohistochemistry shows: VEGF mainly expressed in vascular endothelial cells and heart muscle have strong positive expression, RCS group expression in endothelial within stronger than the normal control group and ACS groups.Conclusions:1 Rosuvastatin may reduce blood pressure in H-hypertension rats.2 To treat rosuvastatin befour acute coronary syndrome, VEGF expression is up regulated in the blood vessels.It may be relationship with established vascular collateral circulation.3 Give rosuvastatin for a long time, in rats of acute coronary syndrome, the asymmetric dimethylarginine expression is lower than the non-drug treatment group. It suggests that giving drug treatment may reduce the risk of acute coronary syndrome. |
