Objective The purpose of this study was to analyze the Indoleamine 2,3-dioxygenasel expression and the lymphocyte subpoplulations distributions as well as the absolute count of regulatory T cells in patients with different subtypes or IPSS risk groups of myelodysplastic syndromes.Methods Real-Time PCR was adopted to examine the IDO1mRNA expression of bone marrow and Flow cytometry was used to measure the percentage of lymphocyte subpoplulations and regulatory T cells in peripheral blood among 27 MDS patiens and 7 healthy volunteers. Meanwhile, the number of lymphocytes in the peripheral blood of each patients and healthy volunteers was to analyzed by blood cells analyzer for further calculated the absolute count of regulatory T cells.Results 1.Compared with those of healthy controls, the RQ of IDO1mRNA was between 2.57±0.12 to 5.01±0.31 and 3.38±0.88 to 7.41±4.48 in each subtype and IPSS risk group of MDS patients respectively, and the differences was statistically significant(RQ>2).Furthermore,the expression of IDO1mRNA have increasing tendency along with the progression of the disease and increasing of IPSS scores, but the differences among each subtype and IPSS risk groups were not statistically significant(1.03<RQ<2.01).2.The percentage of CD3+CD4+T cells,CD3+CD8+T cells, CD16+CD56+natural killer cells and CD4/CD8 ratio have different levels of differences between each subtype or IPSS risk group with healthy controls. In addition, these lymphocyte subpopulations ratios have increasing or reducing tendency with the progression of the disease and increasing of IPSS scores.3. The absolute count of regulatory T cells was between 3.35 to 28.35±2.68 (×106/L) and 4.36±1.33 to 28.35±2.68(×106/L) in each subtype and IPSS risk group of MDS patients respectively, and with the progression of the disease and increasing of IPSS scores,the absolute count of regulatory T cells have increasing tendency and the differences among each subtypes or IPSS risk groups were statistically significant(P<0.05).Conclusion The IDO1 expression and the lymphocyte subpoplulations as well as the absolute count of regulatory T cells can be together for the assessment of MDS patients immune status.Furthermore, these reference indicators also provides some basic experimental data can be utilised to intensive typing and staging for individual treatment and the clinical application of IDO inhibitors in patients with MDS.
