Abstract/Summary:
【Objective】: To detect the changes of subset and function of CD4~+ T cells of myelodysplastic syndromes (MDS), and explore their roles in the mechanism of MDS.【Methods】: Contrast to 11 normal controls, the quantity and ratio of CD3~+CD4~+CD28~+,CD3~+CD4~+CD154~+,CD3~+CD4~+PD-1~+,CD3~+CD4~+CTLA-4~+,CD3~+CD4~+CD25~+ in peripheral blood of 38 MDS patients at prelimilatory phase were detected respectively gated by CD3 and SSC by FACS.【Results】: The percentages of CD3~+CD4~+CD28~+ and CD3~+CD4~+CD154~+ T cells in peripheral blood of MDS were 79.82%±8.99%, 2.33%±1.74%, which CD28~+ was: RA 78.65%±10.4%, RAEB/RAEB-t 80.10%±7.28%, CD154~+ was: RA 1.64%±0.79%, RAEB/RAEB-t 2.76%±2.05% respectively. Those of normal controls were 89.56%±3.06% and 0.20%±0.25%. By comparing with normal controls, though the percentage of CD154~+cells was higher, CD28~+ cells between two groups were significantly low ( t=-5.59 , P﹤0.01). The percentage of CD3~+CD4~+CTLA-4~+T cells of patients with MDS was 2.14%±1.25%(RA 1.51%±0.80% ,RAEB/ RAEB-t 2.65%±1.33%), which was significantly higher than that of normal controls(t=8.66,P﹤0.01); along with progress of disease, the difference between RAEB/RAEB-t and RA was obvious(t=-2.85,P=0.01), especially the ratio of CTLA-4/CD28 which can...
