Objective: Affected by multiple factors, the incidence of breast cancer in women increased day by day. Breast cancer have become the top killer of women in some countries and regions. With the continuous development of medical technology, disease-free survival of patients gradually extended, but local invasion and distant metastasis are still the most serious threat for the patients of breast cancer.The invasion and metastasis of tumor is a complex and dynamic process, including changes in cell adhesion, signal transduction, immune evasion and other aspects. It has been found by study that Disintegrin- metalloproteinase family(ADAMS), EGFR, E-cadherin are closely related to the development of malignant tumors. We detect the expressions of ADAM10, EGFR and E-cadherin in breast cancer tissues and normal breast tissues and investigate the relationship between them and clinicopathological parameters(including patient age, tumor size, clinical stage、histological grade in invasive ductal carcinoma、lymph node metastasis),hoping if we can provide new targets and experimental basis for clinical treatment of breast cancer.Methods:1 Sixty-three specimens of paraffin-embedded human breast cancer tissues and forty specimens of fresh human breast cancer tissues are obtained as experimental group and twenty specimens of normal breast tissues as comparison.All patients didn’t accept chemotherapy, radiation therapy, biological targeted therapy, hormonal therapy before modified radical mastectomy for breast cancer. Normal breast tissues were taken from the tumor edge adjacent that distance was farer than 5cm.2 The expression of ADAM10,EGFR, E-cadherin in normal breast tissues and breast cancer tissues were detected with immunohistochemical technique( paraffin-embedded tissues) and Reverse Transcription-Polymerase Chain Reaction(RT-PCR)(fresh tissues),then the difference of expressions and the relationship between them in breast cancer and clinicopathological parameters were analyzed.3 Statistical analysis: based on an analysis of different objects, Pearson Chi-square(χ2)test, t-test or t ’test of SPSS16.0 software were used for statistical analysis of the results.The probability(P)-value less than 0.05 was considered to be statistically significant.Results:1 Immunohistochemical techniqueThe expression of ADAM10 in breast cancer(68.3%)was higher than that in normal breast tissue(40%).The expression of ADAM10 in the group of histological grade Ⅲ( 88.9%)was higher than that in group of histological gradeⅠ-Ⅱ(60.0%). The expression of ADAM10 in the group of clinical stage Ⅲ( 91.7%)was higher than that in clinical stageⅠ- Ⅱ(60.8%).The expression of ADAM10 in the group of lymph node metastasis(87.9%) was higher than that in the group of lymph node non-metastasis(46.7%). The differences were statistically significant(P<0.05). The differences in the expression of ADAM10 in different age group and different tumor size group were no statistically significant(P>0.05).The expression of EGFR in breast cancer(39.7%) was higher than that in normal breast tissue(15%).The expression of EGFR in the group of Histological grade Ⅲ(66.7%) was higher than that in the group ofⅠ-Ⅱ(27.5%). The expression of EGFR in the group of clinical stageⅢ(75.0%) was higher than that in stageⅠ-Ⅱ(31.4%). The expression of EGFR in the group of lymph node metastasis(51.5%) was higher than that in the group of lymph node non-metastasis(26.7%). The differences were statistically significant(P<0.05). The differences in the expression of ADAM10 in different age group and different tumor size group were no statistically significant(P>0.05).The expression of E-cadherin in breast cancer tissues(66.7%) was lower than that in normal breast tissues(90%). The expression of E-cadherin in the group of histological grade Ⅲ(44.4%) was lower than that in the group of histological gradeⅠ-Ⅱ(82.5%). The expression of E-cadherin in the group of clinical stage Ⅲ(33.3%) was lower than that in the group of clinical stageⅠ-Ⅱ(74.5%). The expression of E-cadherin in the group of lymph node metastasis(54.5%)was lower than that in the group of lymph node non-metastasis(80.0%). The differences were statistically significant(P< 0.05). The differences in the expression of ADAM10 in different age group and different tumor size group were no statistically significant(P>0.05).2 Reverse Transcription-Polymerase Chain Reaction(RT-PCR) methodThe expressions of m RNA of ADAM10, EGFR in breast cancer were higher than that in the normal breast tissues, the expression of m RNA of E-cadherin in breast cancer was lower than that in the normal breast tissues, the differences were statistically significant(P<0.05). The expressions of them were all associated with histological grade, clinical stage and lymph node metastasis concerned. The differences were all statistically significant(P< 0.05). The differences in the expression of ADAM10 in different age group and different tumor size group were no statistically significant(P>0.05).Conclusions:1 ADAM10、EGFR、E-cadherin may be involved in local invasion and distant metastasis of breast cancer.2 The expressions of ADAM10、EGFR、E-cadherin in breast cancer are associated with histological grade, clinical stage and lymph node metastasis concerned. It is expected to provide a new direction for biological targeted therapy and prognosis of breast cancer.
