Objective:To detection the expression of tumor suppressor gene DAB2 interaction protein(DAB2IP)and transcription factors zinc finger E-box binding homeobox 1( ZEB1) in gastric cancer and adjacent normal tissues,and analyze the relationship between clinicopathological parameters of gastric cancer,and preliminary study their correlation and their functions in the development of gastric cancer.Materials and Methods : Random collection of First Affiliated Hospital of Bengbu Medical College,from January 2012 to June 2013,80 cases of gastric surgery archival paraffin blocks for the study,and take one 30 adjacent normal tissue as a control group,using immunohistochemical method for detection of DAB2 IP and ZEB1 in gastric carcinoma and adjacent normal tissue of the group expression,and the expression of both were statistically analyzed, determine whether the difference has statistical significance. Further analysis of the expression of both the patient’s age,gender,histological grade, clinical stage,lymph node metastasis and invasion depth of relationship between parameters of clinical pathology in gastric cancer,and relevance of the expression of DAB2 IP and ZEB1 in gastric carcinoma.Results:1.DAB2 IP in gastric tissue expression was 41.25%(33/80),the positive rate was significantly lower than the adjacent normal tissues 93.33%(28/30),the difference was statistically significant(P <0.005);DAB2IP expression in lymph node metastasis group was 32.00%(16/50),than those without lymph node metastasis positive expression rate of 56.67%(17/30) is lower,the difference between the two groups was statistically significant(P <0.05); with higher gastric cancer grade,clinical stage,the higher the degree of infiltration deeper DAB2 IP lower expression rate,and the difference was statistically significant(P <0.05),while DAB2 IP expression unrelated with age,gender(P> 0.05).2. ZEB1 positive rate in gastric carcinoma 65.00%(52/80),the positive rate was significantly higher than in adjacent normal tissues 10.00%(3/30),the difference was statistically significant(P <0.005); ZEB1 expression in lymph node metastasis group was 78.00%(39/50),than those without metastasis positive expression rate of 43.33%(13/30) is higher,the difference between the two groups was statistically significant(P<0.05). With higher gastric cancer grade,clinical stage higher infiltration deeper level,the higher the positive expression of ZEB1,the difference was statistically significant(P <0.05),ZEB1 expression unrelated with age,gender(P> 0.05).3.In gastric cancer tissues,the positive expression of DAB2 IP was negatively related with the positive expression of ZEB1(r=-0.91),have statistical significance(P<0.05).Conclusions:1.In gastric carcinoma,DAB2 IP expression was downregulated and ZEB1 expression was upregulatied.suggesting that DAB2 IP and ZEB1 expression is closely related to the proliferation,invasion and metastasis of gastric cancer.2.In gastric cancer,the expression of DAB2 IP and ZEB1 was negatively correlated,ut the exact mechanism remains to be further studied;3.The combined detection of DAB2 IP and ZEB1 expression in gastric cancer is helpful to evaluating the invasion and metastasis of gastric cancer and prognosis.
