Objective:DAB2IP is a novel member of tumor suppressor gene, it relates to the invasion and metastasis of various tumors. This study regard DAB2IP as the target, detecting the expression of DAB2IP in normal Colorectal tissues and Colorectal cancer from two levels of protein and mRNA, to explore the relationship between DAB2IP and Colorectal cancer from its occurrence, development, invasion and metastasis. Than to offer a theoretical foundation for clinical to find and cure the tumor early.Methods:Detecting the expression of DAB2IP in 36 cases of normal Colorectal tissues and 127 cases of Colorectal cancer tissues from protein and mRNA levels by Immunohistochemistry(IHC) and nucleic acid In Situ Hybridization(ISH), than using statistical analysis software to analysis the relationship between DAB2IP and the clinical parameters of Colorectal cancer, and finally draw the conclusion.Results:1.The expression of the DAB2IP protein in Colorectal cancer tissues.Experimental result shows that the expression of the DAB2IP protein in Colorectal cancer tissues have no statistical difference between the age, gender, location and the size of the tumor (all the Probability numbers are exceeding 0.05). However, the expression of DAB2IP protein in Colorectal Cancer tissue is down-regulation when compare to the normal Colorectal tissues, both expression has significant difference(Z=-4.147,P<0.05); As different differentiation degree they are, as different in the expression of DAB2IP protein level, From high to low differentiation, the rate of DAB2IP protein expression is down-regulation generally, and all of the expression has significant difference(H=9.795,P<0.05). When compared lymph node metastasis cancer group with no lymph node metastasis cancer group, the expression of DAB2IP protein is lower in former group than the latter group, and both expression has significant difference (Z=-3.552,P<0.05); In the staging of Duke’s, the expression of the DAB2IP protein in stage A and B, C and D stages were not significant difference, but when compared the period of A+B (early-stage) and the period of C+D (late-stage) in the expression of DAB2IP protein, it has significant difference (H=13.161,P<0.05).2.The expression of the DAB2IP mRNA in Colorectal cancer tissuesSame as the group of IHC, the expression of the DAB2IP mRNA in Colorectal cancer tissues has no statistical difference between in the age, gender, location and the size of the tumor(all the Probability numbers are exceeding 0.05); Then the expression of DAB2IP mRNA in normal Colorectal tissues was significantly higher than Colorectal cancer tissue(Z=-5.705,P<0.05); The expression of DAB2IP mRNA in low grade tumor has significant different,when compared with the high grade tumor(Z=-2.238,P<0.05). The experssion of DAB2IP mRNA in Colotretal cancer with lymph node metastasis group is lower than the group that without lymph node metastasis, both expression has significant different(Z=-2.529,P<0.05); In the Duke’s stages, A+B staging also has significant different, when compare with C+D staging in the expression of DAB2IP mRNA (Z=-3.792,P<0.05).Conclusions:1.Both the expression of DAB2IP protein and mRNA are correlation with the occurrence and development for Colotretal cancer.2.Both the expression of DAB2IP protein and mRNA are negatively correlation with the metastasis for Colorectal cancer.3.Both the expression of DAB2IP protein and mRNA are closely correlation with the histologic grading and Duke’s stages for Colorectal cancer.4.Both the expression of DAB2IP protein and mRNA have no correlation with the age, gender, location and the size for Colorectal cancer.5.DAB2IP can be used as an important potential index,to evaluate whether the Colorectal cancer equipped with metastasis potential and prognosis.
