Objectives: In order to find out effective treatment methods for atherosclerosis in Northeast Sichuan,we screened effect of enzyme protein on Hcy metabolism greater levels of normal and coronary heart disease in the population,and to investigate the correlation between Hhcy and coronary heart disease from the gene level.Methods: The work got the approval of the ethics committee of North Sichuan Medical College and the patient’s informed consent,then randomly selected 260 cases who were in hospital from September 2013 to June 2014, and 300 cases of healthy people(normal control group), who were in the clinical symptoms, laboratory examinated, coronary angiography confirmed or ever been diagnosed with coronary heart disease(CHD) patients and readmission to the hospital the outpatient medical health, following the principle of informed consent extracted venous blood. By using the kit to extract peripheral white blood cell genomic DNA, polymerase chain reaction restriction fragment length polymorphism(PCR-RFLP) method to detect the MTHFR polymorphism of C677 T gene, and polymerase chain reaction(PCR) method to detect the CBS 844ins68 gene polymorphism. Meanwhile compare the difference in plasma Hcy level in CHD group and non CHD group, and compare the level of plasma Hcy between the gene polymorphism of each genotype difference. The experimental results using SPSS13.0 statistical software for data analysis.Results:1. Plasma Hcy in CHD group was(18.42±6.8)μmol/l,while in CHD group that was(14.3 ± 4.6) μmol/l,which was significantly lower than the CHD group(P < 0.05).2. The MTHFR C677 T genotype can be divided into three types: CC, CT and TT, three genotypes in CHD group were 20.4%, 43.7%, 35.9% respectively, in normal control group were 39.8%, 40.8%, 19.4% respectively, and the gene frequency differences between groups were statistically significant(P < 0.05). The genotype of CC plasma Hcy concentration was(15.6 ± 3.5) mol/L, CT genotype was(17.5 ±4.7) mol/L and TT genotype was(19.6 ± 7.6) mol/L respectively. The plasma concentration of Hcy(19.6 ± 7.6) mol/L, there were statistically significant differences in plasma Hcy concentration of three genotypes(P<0.01), the level of plasma Hcy in CT and TT were significantly higher than that of CC(P<0.01).3.CBS 844 ins68 genotype including wild type D/D, D/I hybrid type, and I/I mutation type, patient group distribution frequency was 90.38% D/D, D/I 7.69% and I/I 1.92%,and in normal control group 92.67% D/D, D/I 5.33% and I/I 2.0%, so there was no obvious difference statistically significant(P > 0.05). Plasma Hcy levels for the D/D genotype was(17.25±3.62) μmol/l, D/I genotype was(15.74±2.32) μmol/l, I/I genotype was(16.63±2.43) μmol/l, the overall differences of the three genotype corresponding plasma Hcy levels had no statistical significance(P > 0.05).Conclusion:1.For people in northeast Sichuan region, Homocysteine level was significantly related with coronary heart disease, which shows that Hhcy is an independent risk factor for coronary heart disease in northeast Sichuan.2. For the crowd in the northeast region of Sichuan, the MTHFR gene C677 T polymorphism and plasma homocysteine levels were positively correlated, which was different from the the domestic and foreign research.,and show that MTHFR gene C677 T polymorphism may be a very important part in the pathogenesis of CHD risk factors in northeast Sichuan.3. For people in the northeast region of Sichuan, CBS 844 ins68 gene polymorphisms and plasma Homocysteine levels have no obvious correlation, CBS 844 ins68 gene polymorphism is not directly related to the factors of the onset of coronary heart disease, so it may not be the population genetic risk factor for coronary heart disease of people from northeast Sichuan.
