ObjectivesThe purpose of this object is to observe how the MT and HA change. Using the Zuogui Wan and Yougui Wan to harmonic Yin and Yang, to explore the may change rule of sleep substances in the brain and the plasma, in order to offer reasons for clinical treatment of sleep disorders and support the basic research about using the yin-yang sleeping and waking theory to guide the clinical medicine.Methods1.Build mold, grouping and intervention:56 healthy adult male SD rats (260g-320g) were divided into normal control group (n= 8),insomnia group (n= 24),the more sleep group (n= 24).The rat of insomnia group injection of the WAY 100635,the rat of more sleep group injection of the 8-OH-DPAT,all rats injection 3 days. The rat of insomnia group were divided into insomnia control group(n=8), insomnia+Traditional Chinese medicine lavage group (n=8),insomnia +saline lavage group(n=8).The more sleep group were divided into more sleep control group(n=8),more sleep+Traditional Chinese medicine gavage group(n=8),more sleep+saline gavage group(n=8).Insomnia+Traditional Chinese medicine gavage group giving Zuogui Wan, more sleep+Traditional Chinese medicine gavage group giving Yougui Wan, insomnia +saline gavage group and more sleep+saline gavage group giving saline, gavage for 7 days.2. Material:In the 4 days after building abdominal aortic blood then beheaded kill on the ice of normal control group, insomnia control group and more sleep control group. In the 8 days after garaging abdominal aortic blood then beheaded kill on the ice of the rest of the group. Using the high performance liquid chromatography detect of the content of MT and HA in the brain tissue and the plasma.Results1. After the treatment, the dorsal raphe nucleus insomnia group of rats caused by fast REM sleep decreases, awakening time extending; more sleep group of rats caused by fast increased REM sleep, awakening time reduced.2.Compared with the normal control group, the content of HA in insomnia control group is increased, MT content is decreased (P<0.05), the dominant excitatory substances in the brain tissue; insomnia+Zuogui Wan lavage group compared with saline lavage group, the content of HA is decreased, MT is increased (P<0.05).3.More sleep control group compared with the normal control group, the content of HA is decreased, and the MT is increased (P<0.05), inhibitory substances predominate in the brain tissue; more sleep+ Yougui Wan lavage group rats compared with saline lavage group, the content of HA is increased, MT is decreased(P<0.05).4. Compared with the normal control group, the content of HA in insomnia control group is increased, MT content is decreased (P<0.05), the dominant excitatory substances in the plasma; insomnia +Zuogui Wan lavage group compared with saline lavage group, the content of HA is decreased, MT is increased (P<0.05).5. More sleep control group compared with the normal control group, the content of HA is decreased, and the MT is increased (P<0.05), inhibitory substances predominate in the plasma; more sleep+ Yougui Wan lavage group rats compared with saline lavage group, the content of HA is increased, MT is decreased(P<0.05). Conclusion1.Injection of the WAY100635 induced the brain tissue inhibitory neurotransmitter MT decreasing and HA increasing; on the contrary, injection of the 8-OH-DPAT inhibitory neurotransmitter MT increasing and HA decreasing.2. Excitatory and inhibitory substances affect the cycle of sleep, both in a state of mutual restriction. Normal sleep is the transformation of "Wu"and "Mei".3.Zuogui Wan reduces the contents of excitatory substance, Yougui Wan reduces the content of inhibitory substance, eventually keep the balance. Yin-Yang sleeping and waking theory to guide the Clinical medicine for sleep disorders has certainly effect.
