BackgroundThyroid cancer is one of the most common malignant tumor in head and neck, accounting for human malignant tumor 2%, nearly 30 years, the incidence of thyroid cancer in the global scope increased. Papillary thyroid carcinoma(PTC)is the most common in thyroid cancer, however, there is no specific tumor marker, assist in the diagnosis of thyroid papillary carcinoma. Tumor development is a pathological process of many genes, multi stage and multi factors, activation of oncogene and tumor suppressor which loss of function gene is the molecular basis of cancer cancer. Thyroid cancer related gene 1(TC-1) is located in chromosome 8 open reading frame is a highly conserved sequence of a gene, its coded TC-1 nuclear protein belongs to natural folding protein family. TC-1 as a positive regulatory factor involved in the Wnt/β-catenin beta catenin signaling pathways regulating, and closely related and metastasis of tumor proliferation, invasion. TC-1 has aroused the concern of many scholars, detection in the lung cancer, ovarian cancer, gastric cancer in the expression of, although earlier found in thyroid tumors, but the study of its mechanism is less, between the occurrence and development, and metastasis of thyroid tumor relationship reported. This study used immunohistochemistry, real-time fluorescent quantitative reverse transcription polymerase chain reaction(q PCR), Western blot detection of thyroid papillary carcinoma and their adjacent tissues(NCE), metastatic lymph nodes,the expression level of TC-1 in nodular goiter, and to explore the correlation between the expression of TC-1 in papillary thyroid carcinoma and its occurrence, development. ObjectiveUsing real-time fluorescent quantitative reverse transcription polymerase chain reaction(q PCR), immunohistochemical method, Western blot detection in patients with nodular goiter, tumor tissues and their adjacent tissues(NCE), metastatic lymph nodes in the expression of TC-1 and analysis of its relevance, to investigate the correlation between protein expression and thyroid cancer, the relationship of development and cell invasion and metastasis behavior, as molecular papillary thyroid cancer specific biomarkers feasible or not, and provides a new idea for the targeted therapy of thyroid cancer. MethodThe specimens were taken from patients with thyroid tumors in the First Affiliated Hospital of Zhengzhou University from 2013 September to 2014 February resection operation. All patients were not receiving preoperative chemotherapy, radiotherapy and other treatment, and the specimens were confirmed by pathology, all specimens of formalin fixed, paraffin embedded. The expression of TC-1 was detected by immunohistochemical method in thyroid tumor tissue, using real-time fluorescent quantitative reverse transcription polymerase chain reaction(q PCR), papillary thyroid cancer lymph node tissue, detection of Western expression level in blot node metastases of TC-1 gene m RNA and protein, and statistical methods of correlation analysis using SPSS 17 software. Results1.q PCR reaction revealed, expression of m RNA in PTC were significantly higher than the corresponding para carcinoma tissues, and PTC TNM staging, pathological grading and lymph node metastasis(P<0.05).2.Immunohistochemistry showed the positive expression of TC-1 was mainly located in cytoplasm, positive performance for the brown. There were statistically significant differences in TC-1 among groups(P<0.05), and the expression of tissue and the corresponding adjacent difference was significant(P<0.05), and PTC TNM staging, pathological grading and lymph node metastasis(P<0.05). Results showed that Western blot, TC-1 protein in integral optical density value of PTC is(2546±195) was significantly higher than that of the corresponding para cancer tissues(992±75)(P<0.05).Comparison of TC-1 between different types of thyroid cancer, and PTC TNM staging, pathological grading and lymph node metastasis(P<0.05).3.TC-1 gene m RNA and protein expression of correlation, TC-1 gene m RNA expression increased mostly with TC-1 protein expression increased, with correlation(P < 0.05). Conclusions1.High expression of TC-1 in papillary thyroid carcinoma and papillary thyroid carcinoma and TNM stage, pathological grade and lymph node metastasis was significantly related to may with papillary thyroid carcinoma occurrence and development.2. TC-1 can be used for early diagnosis, screening of thyroid papillary carcinoma, for evaluation of thyroid papillary carcinoma metastasis and prognosis has certain value.
