Inhibitory Effect Of Baicalein On Gastric Cancer Resistant Cells SGC7901DDP Via The Modulation Of Notch 1 And IncRNA Ak022798

ObjectiveTo realize the effect of baicalein on growth inhibition and apoptosis in SGC7901 DDP cells; to investigate the mechanism between baicalein and the expression of Notch 1 and lnc RNA AK022798.MethodsCCK-8 method was used to assay the cell proliferation; Flow cytometry combined with Annexin V-PI double staining was used to determine cell apoptosis rate; PCR array was used to screen the related signaling molecules; Westernblot blotting was used to detect the expression of protein; Quantitative RT-PCR was usd to detect m RNA; si RNA was designed to inhibit RNA transcriptional level. DPS software was used in the statistics, Student-t test was used in comparison between two groups(P<0.05 and 0.01, significant difference).Results1. Baicalin inhibited the growth of SGC7901 DDP cells and induced its apoptosis.CCK-8 assay showed that baicalein decreased the OD value in SGC7901 DDP cells(P< 0.01). Flow cytometry analysis showed that baicalein increased the apoptosis rate of SGC7901 DDP cells.2. Baicalein downregulaed Notch1 m RNA.PCR array detection showed that Notch1 gene was downregulated mostly after baicalein treatment.3. Baicalein inhibited the expression of Notch1 protein.Westernblot results showed that baicalein with concentration of 50, 100μg/m L) after24, 72 hours significantly decreased the expression of Notch1 protein.4. Notch1 upregulated lnc RNA ak022798.The bioinformatics analysis results showed that Notch1 decreased the levels of ak022798 in lnc RNA.5. Decreased Notch1 through ak022798 was involved in baicalein inhibory effect.Immunofluorescence and CCK-8 results showed that ak022798 inhibition through si RNA in SGC7901 DDP increased cell proliferation significantly. Compared with the control, cell apoptosis rate decreased significantly(P< 0.01).Conclusion1. Baicalin inhibits the growth of SGC7901 DDP cells and induces its apoptosis.2. Effect of baicalein is related to inhibition of Notch 1 and upregulation of lnc RNA AK022798.
