Objective:To explore the role of S100A16 in depression and to reveal more molecular mechanisms of depression and more function of the protein S100A16. Methods:30 adult male B6 mice were Chose, randomly divided into two groups, the normal control group and the depression group, each group only 15. We adopt the separation and chronic unpredictable mild stress (CMS) to preparation the depression mice. Body weight and fur score were observed weekly. The effectiveness of the proposed depression model were evaluated after preparation about 5 weeks. Enzyme linked immunosorbent assay(Elisa) were used to observe the changes of the S100A16 protein in mice serum when the depression model were confirmed to establish successful. Western blot were used to detect the expression of S100A16 and related proteins in the mice hippocampus. We collected the 42 cases of depression patients and 55 normal serum, to observe and analysis the level of S100A16 in human serum.Results:(1) After used the separation and chronic unpredictable mild stimulation 5 weeks, we found that the depression group mice’s weight drop to 17.488±1.08g from 21±0.69g, working dilatorily and filthy fur. In the forced swimming test (FST), the time of non-swimming increased from 59.4±25.65sec. to 113.1±28.47sec. and the sucrose preference test were positive. The level of S100A16 in the Depression mice serum 3.15±2.19 ng/ml is lower than the normal control group 6.13±2.68 ng/ml with significant statistical significance (p< 0.001). (2) Depression symptom were assessed with the Hamilton Depression Rating Scale (HAMD). To compare and analyze the blood fat in different groups the depression patients and the normal health people. We found that to compare with normal people the total cholesterol and high-density lipoprotein cholesterol declined in depression patients, and triglyceride increased. There was significant difference between the Depression patients serum 3.15±2.19 ng/ml and normal contrast group 6.13±2.68 ng/ml in the level of S100A16 protein(p<0.05).Conclusion:Protein S100A16 is closely related to the depression which induced by chronic mild stress. The mechanisms of those effects still need more research.
