The dried heartwood of Caesalpinia Sappan L. which is called Sappan Lignum belongs to the family of Leguminosae. The Chinese Pharmacopoeia which published in 2010 records it has the effect of removing blood stasis and detumescence, etc. Sappan Lignum also has anti-virus found in modern scholars domestic and overseas, such as immunosuppression and anti -tumor effect. Today by scholars at home and abroad confirm that from Sappan L were isolated compounds with a variety of active ingredients, this may be the reason why Sappan Lignum has the beneficial effects of the material basis for certain diseases. Therefore, it’s important to screening anti-tumor components analysis of the chemical composition for Sappan L. medicinal importance.The research in this paper screens the dried heartwood of Sappan L.. On the basis of preliminary work in the laboratory, we examine the T24 cells killing effects of Sappan L.. Besides, we analyze the resulting product combined with HPLC-MS/MS technology. So we provide a new method for identification of the chemical constituents of the aqueous extract from Sappan Lignum. Both provided the basis for studying the medicinal function of brazilwood in the future.The main research contents and conclusions are as follows:1、By the study of anti-T24 cells using aqueous extract of Sappan L.,we analysis the correlation between ratio of cells killing and content of main component. The result shows that the antitumor composition in Sappan L. is brazilin. Besides, the pure brazilin obtained through separation by column chromatography. It’s content reaches 99%, and the chemical structure of the pure is characterized by spectral data.2、This experiment has been a simple and effective brazilwood extraction process for active ingredients. The content of active ingredients in the product reaches 50% at last. This process is a simple, low-cost, pollution-free, scientific and feasible technology. And it’s easy to use it in industrial production.3、HPLC-MS/MS has been used in the analysis of the chemical constituents of aqueous extract from Sappan Lignum. Confronted by spectral data for resolution and with reference to related literature, we have obtained 16 compounds.The main ingredients are quercetin, brazilin, protosappanin B, etc.Our analysis provides new methods for the constituents of Sappan L., laying the foundation for their correlation with the efficacy of chemical constituents, and provide a basis for studying the anticancer effects of brazilwood in the future.4、This article establishes a method for detecting residual solvent extracts of brazilwood powder. The method in ethyl acetate and ethanol concentration range of 0.1~1.0 mg/mL good linear relationship. The ethyl acetate and ethanol RSD were 2.4% and 3.1% in precision experiments, and the mean recovery of ethyl acetate and ethanol is 101.4% and 100.3% respectively. This method is sensitive, reproducible and can be used for quality control in actual production. In addition, we also tried to get a simple and efficient, and can be applied to industrial production methods of removing residual solvent extracts of brazilwood powder.
