Objective: To investigate effect of Ping Chuan Ning decoction on expression of E GF、ERK2、p-ERK2、MEK2、C-JUN、RAF、C-JUNm RNA in lung tissues of a sthma rats. To explore the molecular mechanisms of prevention and treatment of asthma with Ping Chuan Ning decoction. To prove the importance of Ping Chua n Ning decoction in prevention in asthma airway remodeling.Methods: Totally 140 male SD rats were randomized into 7 groups,20 in each g roup.To copy the ovalbumin sensitized in rat asthma model.To sensitize rats by using 1ml suspension with intraperitoneal injection of 10% egg protein,aluminum h ydroxide, physiological saline at the first, eighth days.At the fifteenth day, sed1% o valbumin solution ult RASonic atomization and gave the cold stimulation for 7 cons ecutive days, 30 min enery time, one time every day.Modeled after 21 days,normal,model control groups were given the sanme amount of distillde water every day.the dexamethasone group,guilongkechuanning group and high、medium、low dose groups of Ping Chuan Ning decoction were given each drug drench according to h uman and rat body surface area than the conversion operator.After 4 weeks disse ctiong the rats and taking out the lung organization.The lung histopathology chan ges were observed by HE staining.Immunohistochemistry method was used to ob serve remodeling in the lungs’ small and medium airway. Expression of C-JUNm RNA in the lungs at m RNA levels were detected by RT-PCR. Wersten blot met hod is used to observe expression of EGF、ERK2、p-ERK2、MEK2、C-JUN、R AF in the lung tissue.Results: HE staining results show that compared with the normal group, model group had obvious airway remodeling phenomenon; Compared with model group, the airway remodeling in each treatment group were properly inhabited. Found by Western Blot method that compared with normal group, EGF in pingchuannin g large dose grop has no specific change(P > 0.05), other groups of EGF prot ein expression were higher(P < 0.01); RAF in model group and Gui Long group,the protein expression have increased(P < 0.01, P < 0.05), the rest groups haveno obvious change; MEK2 in big and middle dose groups without specific chan ges(P > 0.05), other groups of protein expression were significantly increased(P < 0.01); P- ERK2 and ERK2 in model group, Gui Long group,pingchuanning low does groupand dexamethasone group were increased(P < 0.01, P < 0.05), and other groups have no statistical significance; C- JUN in pingchuanning mid dle dose groups has no specific changes(P > 0.05), other groups of protein exp ression were higher(P < 0.01); And compared with model group, the RAF, ME K2, ERK2, C- JUN in addition to Gui Long, dexamethasone and pingchuanning low dose groups were decreased(P < 0.01, P < 0.05), but the other groups hav e no statistical significance; EGF except Gui Long group without statistical signifi cance, the expression of proteins in the other groups were decreased(P < 0.01, P < 0.05); P- ERK2 except Gui Long and pingchuanning low doe groups have no statistical significance, the expression of proteins in the other groups were de creased(P < 0.01, P < 0.05); C- JUN except for pingchuanning middle dose g roup has no specific change(P > 0.05), the expression of proteins in the other groups were decreased(P < 0.01, P < 0.05). Through Sq RT-PCR detection: com pared with normal group, each group C- JUNm RNA expression were increased(P < 0.01); And compared with model group, in addition to pingchuaning large and middle dose groups were decreased(P < 0.01), the other groups have no st atistical significance.Conclusion: Pingchuanning large and middle doses can reduce the cold-asthmaed rats’ EGF, ERK2, P- ERK2, MEK2, MEK2, C- JUN, RAF, C-JUNm RNA le vels in lung tissue, alleviate airway inflammation of cold-asthmaed rats, delay th e airway remodeling and cure the cold asthma.
