Tripterygium wilfordii, the dry root and rhizome of Celastraceae Tripterygium,was proved to anti-inflammatory, anti-neoplasm, regulate the function of immunity,anti-procreation, and so on, which was applied for the refractory disease treatment,such as rheumatoid arthritis, chronic nephritis, and widely used in the clinical.However, a series of toxic and side effect was emerging with the increased use and deeply study, hepatotoxicity, renal toxicity, and so on, and the clinical application was certainly restricted. Confined the toxicity furthest, and remained or increased the curative effect has become a clinical issue that needs to be addressed urgently.Composed by astragalus membranaceus, semen coicis, tripterygium wilfordii, and centipede, Xinfeng capsule was proved to obtaine a significant effect on the rheumatoid arthritis treatment, which was a useful dosage and suitable for patient,with no tripterygium wilfordii-toxicity, despite the toxic of tripterygium wilfordii existed. Decrease the toxic of tripterygium wilfordii by compatibility of medicines is an important countermeasure, so investigate the toxic difference of each compatibility group on the main target organ of rat is researched in this study by using the theory of compatibility to reduce the poison and decomposing the recipe test, which aimed at filtrating the best compatibility group to decrease the toxic, and understanding the new method and idea to decrease the toxicity by investigating the key component change before and after drug combination, and increasing the clinical safety.Objective:(1) Establish the decomposed and composed method of small complex prescription of tripterygium wilfordii.(2) Select the best combinations group to decrease the toxic of tripterygium wilfordii.(3) Investigate the material base changes of best combinations group(1) Three factors and two-levels of L8(23)orthogonal experiment is applied in the Xinfeng capsule and the decomposed and composed method;(2) Collect the blood, heart, liver, kidney, and ovary(testicle) of rat by modern pharmacological options;(3) Detect the serum biochemical index of rat in each group by Elisa and compare the effect of each group on the blood, heart, liver, kidney, and ovary(testicle).Cardiac function indexes: creatine kinase(CK), Creatine Kinase MB(CK-KB); liver function indexes: Alanine transaminase(ALT), Aspartate transaminase(AST); kidney function indexes: creatinine(CREA), Blood urea nitrogen(BUN); genital system function index: female rat: pregnendione(P), follicle-stimulating hormone(FSH),luteinizing hormone(LH); male rat: testosterone(T),(LH).(4) Observe the histomorphology changes of main toxic target organ(heart, liver,kidney, ovary, testicle) after samples handled.(5) Compare the content changes of wilforlide and triptonide in the best recipe by high performance liquid chromatography.Result:(1) compared with the normal group, ALT and AST level in TW group were increased significantly,(P<0.05), suggesting that liver cells may be impaired by TW. Each compatibility group compared with TW group, the level of ALT and AST in Leihuang, Leiyi, Leihuangyi, Leihuangwu, Leiwuyi group were all decreased, which in Leihuang group can significantly reduce th level of two indicators, Leiyi group can significantly decrease the AST level with significant difference(P<0.05).(2) compared with the normal group, the levels of CREA and BUN in TW group and Leiwu group increased significantly(P<0.05). Each compatibility group compared with TW group, the level of CREA and BUN in Leihuang group has a significant decrease,and BUN in Leihuang and Leiyi group all decreased significantly(P<0.05).The rest of the compatibility groups, can reduce CREA and BUN level, but thedifference was not statistically significant(P>0.05).(3) Compared with the blank control group, the level of CK and CK-MB in TW group were increased significantly, with significant difference(P<0.05). Each compatibility group compared with TW group, the two index of Leihuang group decreased significantly(P<0.05). The rest of the compatibility groups in addition to Leiwu group can reduce the level of CK-MB significantly(P<0.05).(4) Compared with the blank control group, Tripterygium wilfordii group can reduce the male rats of the levels of T and LH, and Tripterygium wilfordii group decreased the most obvious, with significant difference(P<0.05). Compared with Tripterygium wilfordii group, Leihuangyi group could significantly increased the level of T, LH(P<0.05), the other group of this two index did not change significantly. Instructions of Astragalus combined with Tripterygium wilfordii can be reduced to a certain degree of male reproductive system toxicity.(5) Compared with the blank control group, Tripterygium wilfordii group can significantly increased the level of FSH and LH and decreased the level of P with significant difference(P<0.05). Compared with TW group, other groups all can reduced the level of P, Increased the level of FSH and LH, all of which the Leiyi group was most obvious, with significant difference(P<0.05).(6) Light microscopy showed that triptolide different compatibility group heart,liver, kidney, testis, ovary histological damage than the control group had varying degrees of ease, integrated various target organs, including the most obvious group Lei Huang.(7) After Astragalus Combination, wilforlide, triptonide levels were lower than Tripterygium wilfordii group.Methods:Conclusion:Based on the toxicity changes of main target organs of Tripterygium wilfordii due to compatibility in Xinfeng capsule, it could be synthetically inferred that the content of Triptolide and Triptonide could be decreased by combination ofT.wilfordii and Astragalus,thus, which provided that Tripterygium wilfordii induced toxicity could be decreased chemical basis when Tripterygium wilfordii and Astragalus were simultaneously used.
