The Study Of Effect Of Bupishen Huoxie Traditional Chinese Medicine On Proliferation And Apoptosis Of The Parathyroid Glands Of Rats With Chronic Renal Failure

ObjectiveBuPishen Huoxie traditional chinese medicine developed by professor Wang Xiaoguang can tonify kidney and spleen, promote blood flow, ascend clear and dispel turbidity in Chronic renal failure. Clinical practice proved that the CRF patients using this medicine can achieve good curative effect.This study observed the effect of BuPishen Huoxie traditional chinese medicine on 5/6-nephrectomized rats combined with high phosphorus diets, which emerged secondary parathyroid gland hyperfunction and calcium, phosphorus (P) metabolic disorder. We studied the effect of the medicine on lowering intact parathyroid hormone(iPTH) secretion and promote eliation, and explored the regulation of parathyroid glands cell apoptosis and proliferation, the prot-ection for renal function, to provide experimental basis for the further application.Methods5/6-nephrectomized (Nx) and sham operated with high phosphorus diets rats were assigned to five groups:(1)Sham-operated group (Sham group, n=10); (2)5/6-nephrectomized group(Nx group, n=10); (3)5/6-nephrectomized+Cal--citriol (Cal group, n=10); (4)5/6-nephrectomized+traditional chinese med--icine group(Tcm group, n=10);(5)5/6-nephrectomized+Calcitriol+traditional Chinese medicine group(Tcm+Cal group,n=10).Serum creatinine (SCr), BUN, P, and iPTH levels were detered at 1 week before operation and day7,14,21,28 after operation. At the end of experiment, four rats of every group were killed and the kidneys, thyroid glands and parathyroid glands complex were removed to be observed. We compared the number of para--thyroid gland cell (N) per unit area and the expression of the apoptotic marker TUNEL by immunohistochemistry.Analyzed the exact effect of Chinese and western medicine combination through the inter-group control between groups of Tcm group, Cal group and Tcm+Cal group to provide theoretical basis for cl-inical application.Results1. After 5/6-nephrectomized (Nx) and high phosphorus diets, the level of SCr, BUN, P, iPTH, N in Nx group were significantly increased than in Sham group (P<0.01). The level of body weight(BW), apoptosis index(AI)were decreased than in Sham group (P<0.05). The residual kidney tissues of Nx group appeared pathological damage.2. In Tcm group and Tcm+Cal group, the level of SCr, BUN were significantly decreased, BW was increased, the residual kidney tissues were protected than in Nx group. The Cal group had no effects on kidney function, residual kidney tissues protection and BW (P>0.05). In three treatment groups, the level of P, iPTH, N were significantly decreased,AI was increased than in Nx group (P (0.01).3. The level of P, N had no significant difference in Tcm group and Cal group(P>0.05). The level of iPTH in Tcm was decreased than Cal group (P< 0.05). The level of AI in Tcm was increased than Cal group (P<0.01). In Tcm+Cal group, the level of P, iPTH, N, AI had significant difference with Cal group (P <0.05).The level of AI in Tcm+Cal group was significantly increased than in Tcm group (P<0.01). ConclusionBuPishen huoxie traditional chinese medicine can decrease the level of serum creatinine, BUN, phosphorus (P), iPTH; inhibit parathyroid gland hyperp--lasia and promote its apoptosis and check the progress of SHPT, which had obvious advantage over Calcitriol.
