Objective:To investigate the expression of miR-34a and miR-125b in the peripheral blood mononuclear cell(PBMCs) of patients with rheumatoid arthritis(RA), Osteoarthritis(OA), to further explore the role of miR-34a and miR-125b in the pathogenesis of RA,to find out the marker which can dagnose RA early,and effective drug treatment targets, reduce the disease morbidity in patients with rheumatoid arthritis, to improve the patient’s quality of life.Methods:There were 35 patients with RA,15 patients with OA and 20 healthy controls as the research object. PBMCs were isolated from peripheral blood in using Lymphocytes Separation Medium, which was used to extract total RNA, the expression levels of miR-34a and miR-125b were measured using real-time RT-PCR,The results combined with the clinic data of patients were analysed.Results:The levels of miR-34a and miR-125b were significantly lower in the PBMCs from RA patients than healthy individuals and OAgroup (P< 0.05).The expression of miR-125b were significant lower between OA groups and healthy individuals (P< 0.05).The expression levels of miR-34a in RA patients were positively correlated to CRP, were negatively correlated to RF, but not to ESR and anti-CCP,The levels of miR-125b were positively correlated to CRP and ESR,were negatively correlated to RF, but not to anti-CCP. The expression levels of miR-34a and miR-125b in OA patients were positively correlated to CRP and ESR, but not to RF and anti-CCP.This suggests the expression of miR-34aand miR-125b may be related with the disease activity of RA and OA.Conclusion:The expression levels of miR-34a and miR-125b were significantly lower in the PBMCs of RA, and were associated with the laboratory indexes of RA, Its expression level might be related to RA occurrence, development and transformation, To explore the role of miR-34a and miR-125b in rheumatoid arthritis and the possible mechanism for help.
